摘  要:目的对羊急性心梗卸负荷模型的micro RNA表达谱进行初步差异分析,探索LVAD卸负荷的调控机制。方法根据micro RNA表达谱筛选出差异表达miRNA,用靶基因预测软件PITA、mi Randa得到差异表达miRNA的靶基因,通过GO数据库和KEGG数据库对靶基因进行GO注释和KEGG pathway分析。结果筛选得到卸负荷组与对照组相同表达miRNA 3个,差异表达miRNA 22个。其中mi R-21、mi R-26a、mi R-30c、mi R-125b与心肌细胞凋亡调控相关,mi R-30a-3p可能与缺氧后心肌细胞自噬相关,mi R-218a、mi R-541-5p可能与肥厚性心肌调节相关,13个miRNA未见与心梗相关的报道。GO和KEGG分析发现差异miRNA靶基因富集的生物功能和信号通路与LVAD卸负荷作用密切相关。结论通过对micro RNA表达谱的差异分析,发现了9个参与卸负荷作用调节和13个未知功能的micro RNA表达,为进一步深入分析急性心梗的调控网络提供了一个新的线索。Objective To analyze the difference of micro RNA expression profile of sheep acute myocardial infarction(AMI) unloading model and explore the regulation mechanism of unloading of left ventricular assist device(LVAD). Methods Differentially expressed mi RNAs were screened out based on the micro RNA expression profile. Target genes for differentially expressed mi RNAs were obtained using target gene prediction software PITA and mi Randa. GO annotation and KEGG pathway analysis were performed on target genes using the GO database and the KEGG database. Results 3 identically expressed mi RNAs and 22 differentially expressed mi RNAs were screened. Among them, mi R-21, mi R-26 a, mi R-30 c, and mi R-125 b were involved in the regulation of cardiomyocyte apoptosis. mi R-30 a-3 p may be associated with hypoxia cardiomyocytes autophagy, and mi R-218 a and mi R-541-5 p may be associated with hypertrophic myocardial regulation. And, 13 mi RNAs were not reported related to MI. GO and KEGG analysis revealed that the differential mi RNA target genes are enriched in biological functions and signaling pathways that are closely related to unloading effect of LVAD. Conclusions By analyzing the differences in micro RNA expression profiles, we found 9 micro RNAs involved in unloading regulation and 13 micro RNAs with unknown functions, which provided a new clue for further analysis of the regulatory network of AMI.
