翡翠贻贝粘附蛋白生物相容性及粘附功能研究

摘  要:海洋贻贝粘附蛋白以其具有很强的粘接力和高度的防水耐腐蚀性能被人们所广泛关注,随着研究的深入发现粘附蛋白可用于水下粘合剂和密封胶的应用,将来在国防和海洋工程方面会有广泛的应用前景。本研究从翡翠贻贝足部直接提取其粘附蛋白(Pernaviridis foot protein,Pvfp),通过酸尿素聚丙烯酰胺电泳检测发现提取的总蛋白中含有七种分子量大小不同的蛋白。多巴试剂染色显示其中四种蛋白多巴含量较高。对比较分析Pvfp在四种不同属性的材料表面粘附量,我们发现Pvfp具有良好的防水性能,且其在金属材料表面比在聚合材上料粘附性更强。利用MTT细胞毒性实验和流式细胞仪检测技术分析显示,Pvfp具有良好的细胞相容性且能促进细胞的增值。以上实验结果为以后翡翠贻贝粘附蛋白在生物易用粘合剂中的应用提供研究基础。Marine mussel adhesive protein has the properties of extremely moist-adhesion,water resistance and adhesion.so it maybe be used to make underwater sealants and adhesives, and also has a wide range of applications in marine engineering and national defense. In my reseach,through the technology of acetone precipitation Pvfp was directly extracted from the Perna viridis foot. Seven kinds of CBBR-positive bands were found in the crude extract by the analysis of the acid-urea-polyacrylamide gel electrophoresis. The quantities of DOPA were high in four of NBT-positive bands. The adhesion experiments on four kinds of materials surface implied that Pvfp has good waterproof, its adhesion effectve on metal materials surface is stronger than that on polymeric materials surface. By the cytotoxicity with MTT and flow cytometry experiments we found that the Pvfp has good cell compatibility and can induce cell proliferation. These would support the evidence of Pvfp application in tissue adhesive.
