摘  要:Bph14基因是第一个克隆的水稻抗褐飞虱主效基因,来自药用野生稻背景的B5品系。本研究通过比对Bph14基因在B5和参考基因组日本晴上核苷酸序列差异,开发了一个功能标记H14,在供体B5和7个受体亲本之间存在明显的多态性,能准确地区分含或者不含抗虫基因Bph14的基因型。利用H14标记对Bph14基因的分离群体进行基因型检测和接虫鉴定,根据标记检测结果与抗虫表型推算出标记的选择效率达95%以上。研究结果表明该标记准确可靠,可为应用水稻抗褐飞虱分子标记辅助育种提供科学依据。The Bph14 gene is the first cloned major gene of rice brown planthopper resistance,and is from B5 strain of medicinal wild rice.A functional marker H14 was developed by comparing the nucleotide sequence difference of Bph14 gene between B5 and reference genome Nipponbare in this study.The marker H14 showed a significant polymorphism between B5 and seven recipient parents,and also could obviously distinguish the genotypes with or without the resistant gene Bph14.The H14 marker was applied for genome detection and insect identification on the segregation population of Bph14 gene.Marker selection efficiency was calculated to over 95% according to the marker detection result and insect-resistant phenotype.The result indicated that the marker was accurate and reliable,which would provide scientific basis for the application of molecular marker-assisted breeding of rice anti-brown planthopper.
