摘 要:为鉴定一次黑山羊养殖场暴发的皮肤癣菌病原,采集患病羊病变皮屑进行病源分离,对分离的真菌进行形态学鉴定和ITS序列分析,并采用CLSI M38-A2微量稀释法筛选敏感天然药物。结果表明:分离的真菌菌落形态、孢子和菌丝结构与疣状毛癣菌相符,其ITS序列(登录号:KX619436)与Pchelin公布的T.verrucosum(登录号:KU378220)同源性最高,达99.99%;18味天然药提取物对分离真菌最低抑菌浓度和最低杀菌浓度试验表明,知母的抗菌效果最好(40 mg/m L,160 mg/m L)。这表明,引起该养殖场黑山羊皮肤癣菌病的病原为疣状毛癣菌,知母为该分离菌的最佳敏感天然药。To identify the causal agent for ringworm in black goat farm,in this study,pathogenic lesions of goat skin were collected for pathogen isolation and ITS sequence analysis. Moreover,M38-A2 CLSI micro dilution method was used to screen the sensitive natural drug. Results showed that colony morphology,and spore and hypha structures are the same as those of Trichophyton verrucosum. The ITS gene sequence of T. verrucosum( accession number: KX619436) had the highest homology(99. 99%) with the published( KU378220). Then,extracts of 18 flavor natural medicines were applied to antifungal test,in the minimal inhibitory concentration( MIC) and minimum fungicide concentration( MFC),and it was shown that the antifungal effect of Anemarrhenaasphodeloides is the best(40 mg/mL,160 mg/mL). Results indicated that the T. verrucosum is the pathogen that caused Dermatomycosis of goat in the farm,and Anemarrhenaasphodeloides is the most sensitive natural medicine in vitro.
