基于网络药理学的“白鲜皮-地肤子”药对治疗湿疹作用机制研究

摘　　要：目的:基于网络药理学,探讨"白鲜皮-地肤子"药对治疗湿疹的作用机制。方法:本研究依托中药系统药理学数据库和分析平台,筛选出"白鲜皮-地肤子"药对的21个化合物并预测靶点,通过治疗靶点数据库、遗传关联数据库预测治疗湿疹的靶点,并通过STRING数据库构建蛋白互作网络;采用Cytoscape软件构建"白鲜皮-地肤子"药对的化合物-预测靶点网络、与湿疹相关的化合物-潜在靶点网络。结果:与湿疹相关的化合物-潜在靶点网络共包含17个靶点,涉及视黄酸受体RXR-α(RXRA)、核受体共激活因子2(NCOA2)、雄激素受体(AR)、转录因子p65(RELA)及肿瘤坏死因子(TNF)等。结论:本研究结果验证和预测了"白鲜皮-地肤子"药对治疗湿疹的作用机制,并为进一步深入揭示其作用机制提供了科学依据。OBJECTIVE： To probe into the mechanism of ＂cortex dictamni-fructus kochiae ＂ in treatment of eczema based on network pharmacology. METHODS： Traditional Chinese Medicine Systems Pharmacology database was used to screen 21 compounds of the herb pair of ＂cortex dictamni-fructus kochiae＂and to predict its target spots,Therapeutic Target Database and Genetic Association Database were used to predict the target spots for the treatment of eczema,and protein-protein interaction（ PPI） network was established on basis of STRING database; Cytoscape software was adopted to construct the networks of compounds-predicted target spots of the herb pair of ＂cortex dictamnifructus kochiae ＂ and compound-potential target spots related to eczema. RESULTS： The network of compoundpotential target spots related to eczema contained 17 target spots,which involved retinoic acid receptor-α（ RXR-α）,steroid receptor coactivator-2,androgen receptor,transcription factor-p56 and tumor necrosis factors. CONCLUSIONS：This research has verified and predicted the mechanism of ＂cortex dictamni-fructus kochiae＂in treatment of eczema,which provides scientific basis for further revealing its mechanism of action.
