摘 要:目的:观察复方黄栌口服液对小鼠黄疸型肝炎模型肝匀浆总胆固醇(total cholesterol,TC)、三酰甘油(triacylglycerol,TG)、丙二醛(malondialdehyde,MDA)及超氧化物歧化酶(superoxide dismutase,SOD)水平的影响。方法:72只小鼠随机分为空白组,模型组,护肝宁片组[1 g·(kg·d)-1],大剂量、中剂量、小剂量复方黄栌口服液组[(30 mL·(kg·d)^(-1)、15mL·(kg·d)^(-1)、7.5 mL·(kg·d)^(-1)],给药方式为每天灌服1次,连续10 d。于第8天给药2 h后,各给药组及模型组均灌服体积分数0.4%异硫氰酸萘酯橄榄油溶液(100 mg·kg-1),空白组给予同体积的橄榄油,48 h后取材,取材前12 h禁食不禁水。将小鼠脱颈椎处死,观察各组小鼠组织病理学变化。另取肝脏,用冷生理盐水制成匀浆,检测肝匀浆MDA、SOD、TC、TG。结果:复方黄栌口服液各剂量组和护肝宁片组TC、TG水平均低于模型组(P<0.01)。复方黄栌口服液各剂量组和护肝宁片组MDA均低于模型组,SOD水平均高于模型组(P<0.01)。组织形态学显示:模型组小鼠肝组织中肝小叶分界不清晰,肝细胞索排列紊乱,出现局灶性坏死、嗜酸性变和点状坏死,有胆栓的形成;护肝宁片组小鼠肝组织中肝小叶分界不清晰,肝细胞出现局灶性坏死及点状坏死,汇管区可见炎细胞侵润,有少量胆栓形成;大剂量复方黄栌口服液组小鼠肝组织中肝小叶分界较清晰,可见部分肝细胞索以中央静脉为中心呈放射状排列,部分排列紊乱;肝细胞出现局灶性坏死和点状坏死,同时可见部分再生的肝细胞,胆栓明显减少。结论:复方黄栌口服液可显著改善异硫氰酸-萘酯致小鼠黄疸型肝炎模型症状,显著降低肝匀浆TC、TG、MDA水平,显著升高SOD水平;可显著减轻肝脏的损伤。Objective: To study the effect of compound Huanglu oral liquid on the level of liver uniform plasma total cholesterol( TC),triacylglycerol( TG),malondialdehyde( MDA) and superoxide dismutase( SOD) in mice with jaundice hepatitis. Methods:Taking 72 mice,they were randomly divided into 6 groups: blank group,model group,Huganning tablets group,large,medium,and small doses of compound Huanglu oral liquid group[( 30 m L·( kg·d)-1,15 m L·( kg·d)-1,7. 5 m L·( kg·d)-1]. The administration method was 1 time per day and 10 d in succession. After the 8 D administration of 2 H,each group and model group were filled with 0. 4 % ISO-naphthyl-naphthyl( ANIT) olive oil solution( 100 mg·kg-1). The blank group gave the same volume of olive oil,and after 48 h,the material was drawn. The former 12 h could not help but fast water. After the mouse was removed from the cervical spine,1 piece of liver tissue was cut from the right lobe of the liver and fixed with 10 % neutral formalin. After dehydration,burial,biopsy,and HE staining,histopathological changes were observed. Another liver,made of cold saline solution to detect liver homogenization MDA,SOD,TC,TG. Results: The levels of TC and TG in each dose group of compound Huanglu oral liquid and those of Huganning tablets group were lower than those of model group( P 〈 0. 01). The MDA of compound Huanglu oral liquid group and Huganning group were lower than those of the model group,and the SOD level was higher than that of the model group( P 〈 0. 01). The histomorphology of the model group showed that the liver lobule in the model group was not clear,the liver cell cord was arranged in disorder,the focal necrosis,eosinophilic and punctate necrosis appeared,showing the formation of the biliary embolus,the liver lobule demarcation of the liver tissue in the mice of the liver in the Huganning tablets group was not clear,the hepatic cells appeared focal necrosis and spot necrosis.
