摘  要:目的观察感染旋毛虫小鼠血清及旋毛虫幼虫抗原对MCF-7细胞生长的抑制作用。方法用旋毛虫感染昆明小鼠,分别于第9、28d从小鼠颈动脉取血,离心、分离旋毛虫成虫感染期血清及旋毛虫幼虫感染期血清。用旋毛虫幼虫抗原和上述感染后血清分别处理MCF-7细胞,通过MTS试验检测MCF-7细胞增殖活力,通过划痕试验检测MCF-7细胞迁移能力,通过Caspase-3/7试验和TUNEL试验检测MCF-7细胞凋亡情况。以正常小鼠血清和正常培养基作为对照组。结果旋毛虫抗原、旋毛虫感染的小鼠血清处理对数生长期MCF-7细胞48h后和对照组比较。正常培养基、幼虫抗原组的细胞A490值分别为1.729和1.493,Caspase3/7活性检测值分别为217 979.500和730 395.70,TUNEL检测MCF-7细胞的平均凋亡数量分别为21和57。正常血清、幼虫期血清、成虫期血清组的细胞A490值分别为1.970、1.828和1.641,Caspase3/7活性检测值分别为137 557.700、209 552.367和152 058.700,TUNEL检测MCF-7细胞的平均凋亡数量分别为34、42和38。划痕实验结果显示幼虫期血清,成虫期血清,幼虫纯化抗原较正常血清"疤痕"愈合时间延长。结论感染不同阶段旋毛虫的小鼠血清及幼虫抗原均可抑制MCF-7细胞活力,诱导细胞凋亡。Objective To observe the inhibitory effect of serum from mice infected with Trichinella spiralis and T.spiralis larval antigens on the growth of human breast cancer MCF-7 cells. Methods Female Kunming mice were randomly divided into 3 groups:two infection groups(both were infected with 300 pre-encapsulated larvae)and an untreated group.After infection,blood was collected from a neck artery on day 9 when larvae were present and on day 28 when adult worms were present.Collected mouse serum was centrifuged and separated.MCF-7 cells were treated with both types of infected serum.MCF-7 cells were also treated with commercial T.spiralis larval antigens.Cell growth was determined with an MTS assay,and cell migration was determined using a scratch assay.Cell apoptosis was determined using a caspase-3/7 assay and a TUNEL assay.Normal mouse serum and normal culture medium served as controls. Results MCF-7 cells in the logarithmic growth stage were treated with T.spiralis antigens and serum from mice infected with T.spiralis for 48 h.Cell growth was compared to that of the controls.A490 was 1.970 for normal serum,1.828 for serum containing larvae,and 1.641 for serum containing adult worms.Caspase3/7 activity was 137 557.700 in normal serum,209 552.367 in serum containing larvae,and 152 058.700 in serum containing adult worms.The average number of apoptotic MCF-7 cells detected with TUNEL was 34 in normal serum,42 in serum containing larvae,and 38 in serum containing adult worms.A490 was 1.729 for normal culture medium and 1.493 for medium containing larval antigens.Caspase3/7 activity was 217 979.500 in normal culture medium and 730 395.70 in medium containing larval antigens.Theaverage number of apoptotic MCF-7 cells detected with the TUNEL assay was 21 in normal culture medium and 57 in medium containing larval antigens.Results of the scratch assay indicated that a "scratch" took longer to heal when cells were exposed to serum containing larvae,serum containing adult worms,or medium containing purified larval an
