摘 要:目的阐明2017年汕头市2型登革流行株的基因型别,为其传播途径的分子流行病学研究提供依据。方法采集2017年汕头市疑似登革热患者血清标本,进行登革热病毒检测,阳性者进行全长E基因PCR扩增,对扩增产物进行序列测定,采用生物信息学软件Sequencher5.1、BioEdit和Mega6.0进行序列的比对分析并构建系统进化树。结果有4例血清标本检出登革热病毒2型核酸,其中2例的标本扩增出E基因全长序列(1 485bp),两者序列相似度100%,判定是由同一株病毒引起的感染。该流行株属于Cosmopolitan基因亚型,与东南亚毒株具有较高相关性。氨基酸序列分析显示该毒株E蛋白毒力位点的氨基酸为E126-E、E383~385-E-P-G、E71-A和E390-S。结论汕头市2017年登革病毒流行株可能为东南亚毒株并发生一定变异,其E蛋白毒力位点的氨基酸为E126-E、E383~385-E-P-G、E71-A和E390-S,属于弱毒株。Objectives To reveal the genotype of the type 2 dengue virus(DENV2)from the City of Shantou in 2017 and to provide a basis for the molecular epidemiology study of the pathways of DENV2 transmission. Methods Blood samples from patients were tested for DENV2,and the E gene was identified in patients who tested positive.The full length of the E gene was amplified using PCR.PCR products were subjected to nucleotide sequencing.The E gene was sequenced and phylogenetic analysis was performed using the bioinformatic software Sequencher5.1,BioEdit,and Mega6.0. Results Four patients tested positive for DENV nucleic acids in the City of Shantou in 2017.The E gene from 2 samples was amplified with PCR and sequenced.The full-length sequence of the E gene(1 485 bp)from both samples was similar.The similarity of the two sequences was 100%,so the infection in the 2 patients was confirmed to be caused by the same viral strain.The strain was classified as the Cosmopolitan genotype,and it was closely related to Southeast Asian strains.Amino acid sequencing indicated that amino acids E126-E,E383-385-E-P-G,E71-A,and E390-S in the E protein were sites responsible for the virulence of the strain. Conclusion DENV2 isolated from Shantou in 2017 may have originated from strains circulating in southeast Asia,and mutations were noted in the strain isolated in 2017.Amino acids in the E protein(E126-E,E383-385-E-P-G,E71-A,and E390-S)were the sites responsible for the strain’s virulence.The identified strain is weakly virulent.
