摘 要:目的探讨炎性因子对人口腔黏膜上皮细胞(OMEC)人β防御素(hBD)-2和hBD-3表达的影响。方法收集人健康口腔黏膜组织,进行消化培养后选用第2代人OMEC。培养48 h后将OMEC分为空白对照组、肿瘤坏死因子-α(TNF-α)组、干扰素-γ(IFN-γ)组、白细胞介素-17(IL-17)组、TNF-α+IFN-γ组、TNF-α+IL-17组和IFN-γ+IL-17组,除空白对照组外加入相应的细胞因子。干预24 h后采用实时荧光定量PCR检测OMEC的hBD-2、hBD-3 mRNA相对表达量,酶联免疫吸附试法检测细胞上清hBD-2和hBD-3的蛋白水平。结果 TNF-α组hBD-2 mRNA的相对表达量、IL-17组的hBD-2及hBD-3 mRNA的相对表达量均高于空白对照组(均P<0.05),TNF-α+IFN-γ组、TNF-α+IL-17组、IFN-γ+IL-17组hBD-2、hBD-3 mRNA的相对表达量及hBD-2蛋白水平均高于相应细胞因子单独干预组(均P<0.05)。结论炎性因子能诱导人口腔黏膜上皮细胞表达hBD-2和hBD-3。Objective To investigate the effect of inflammatory cytokines on the expressions of human beta-defensin( hBD)-2 and hBD-3 in human oral mucosal epithelial cell( OMEC). Methods Oral mucosal tissues of healthy subjects were collected. The second-generation human OMECs were selected after digestion and culture. After 48 hours of culture,the OMECs were divided into blank control group,tumor necrosis factors-α( TNF-α) group,interferon-γ( IFN-γ) group,interleukin-17( IL-17) group,TNF-α + IFN-γ group,TNF-α + IL-17 group and IFN-γ + IL-17 group. Then the corresponding cytokines were added to all groups except the blank control group. After 24 hours of intervention,the relative expression levels of hBD-2 and hBD-3 mRNAs in OMECs were detected by real-time quantitative PCR,and enzyme-linked immunosorbent assay was used to detect the protein levels of hBD-2 and hBD-3 in cell supernatant. Results Compared to the blank control group,the relative expression level of hBD-2 mRNA in the TNF-α group and the relative expression levels of hBD-2 and hBD-3 mRNAs in the IL-17 group were higher( all P〈0. 05). The relative expression levels of hBD-2 and hBD-3 mRNAs and the hBD-2 protein levels in the TNF-α + IFN-γ group,TNF-α + IL-17 group and IFN-γ + IL-17 group were higher than those in the group with corresponding single intervention( all P〈0. 05). Conclusion Inflammatory cytokines can induce the expressions of hBD-2 and hBD-3 in human OMECs.
