摘  要:目的:探讨高效液相色谱法(HPLC)测定金银花炒炭前后芦丁等6种有效成分的含量变化。方法:取适量金银花药材,将其均分为6份,每份200g,270℃炒至焦褐色,炒炭细粉过筛后加入50%甲醇,超声1小时,采用0.45μm滤膜过滤后待检。同时制备对照品溶液。采用HPLC法,色谱柱为Aglient phenyl柱(250mm×4.6mm,5μm),以乙腈(A)-0.5%醋酸(B)溶液流动相进行梯度洗脱操作,以355nm为检测波长,柱温26℃,检测金银花炒炭前后芦丁、咖啡酸、绿原酸、槲皮素、木犀草苷、金丝桃苷等6种有效成分含量变化。结果:芦丁、咖啡酸、绿原酸、槲皮素、木犀草苷、金丝桃苷检测进样量均成良好线性关系,线性范围分别为:0.0022~0.83μg(r=0.9996)、0.0020~0.85μg(r=0.9997)、0.0014~9.73μg(r=0.9996)、0.0021~0.84μg(r=0.9995)、0.0024~0.87μg(r=0.9994)、0.0020~0.93μg(r=0.9998)。稳定性、精密性及重复性试验RSD<2.30%,稳定性、精密性及重复性良好。6种有效成分平均加样回收率分别为97.78%、97.76%、96.88%、98.31%、98.85%、99.54%。与炒炭前比较,炒炭后金银花中芦丁含量明显增加,咖啡酸、绿原酸含量明显下降,槲皮素、木犀草苷、金丝桃苷含量无明显变化。结论:金银花炒炭前后6种有效成分存在一定变化,HPLC法可有效检测金银花6种有效成分,且操作简单、稳定性、精密性及重复性好。Objective: To investigate the determination of six effective ingredients in Honeysuckle before and after charred by HPLC. Methods: Taking appropriate honeysuckle herbs,and divided into 6 copies,each 200 g,270℃ fry until coke brown,scrambled charcoal after screening by adding 50% methanol,ultrasound 1 h,0. 45μm filter to be seized. At the same time,prepared the reference solution. The column was Aglient phenyl column( 250 mm×4. 6 mm,5μm),the mobile phase was acetonitrile( A)-0. 5% acetic acid( B) solution,the detection wavelength was at 355 nm. The contents of rutin,caffeic acid,chlorogenic acid,quercetin and other 6 active ingredients were determined by HPLC. Results: Six kinds of active ingredients test injection volume into a good linear relationship. The linear range was 0. 0022 - 0. 83μg( r = 0. 9996),0. 0020 - 0. 85μg( r = 0. 9997),0. 0014 - 9. 73μg( r = 0. 9996),0. 0021 - 0. 84μg( r = 0. 9995),0. 0024 - 0. 87μg( r = 0. 9994),0. 0020 - 0. 93μg( r =0. 9998). Stability,precision and repeatability test RSD〈2. 30%,stability,precision and repeatability. The average recoveries of the six active ingredients were 97. 78%,97. 76%,96. 88%,98. 31%,98. 85% and 99. 54%. Compared with the former,the content of rutin in honeysuckle increased obviously,the content of caffeic acid and chlorogenic acid decreased obviously,the content of quercetin,luteolin and hyperoside did not change obviously. Conclusion: Before and after the honeysuckle charcoald,6 active ingredients there were a certain change,HPLC method can effectively detect six kinds of effective components of honeysuckle,and the simple operation,stability,precision and repeatability.
