同型半胱氨酸对体外培养的大鼠动脉平滑肌细胞增殖的影响

摘　　要：目的 :观察同型半胱氨酸 (HCY)对大鼠动脉平滑肌细胞 (ASMCS)增殖的影响。方法 :利用体外细胞培养技术 ,通过 MTT比色测定、细胞计数及流式细胞仪对细胞周期的测定 ,观察同型半胱氨酸对大鼠动脉平滑肌细胞增殖的影响。结果 :同型半胱氨酸能促进大鼠动脉平滑肌细胞的增殖 ,呈明显的剂量依赖关系。药物作用两天后 ,0 .2 m M· L-1HCY组与对照组相比 ,细胞数目增加 4 6 .4 8%,MTT比色OD值增加 4 5 .6 9%(P<0 .0 1) ;经细胞周期的测定 ,结果表明 HCY作用后 G0 ～ G1期细胞数由对照组的 87.2 2 %降低至 76 .6 5 %,减少了 12 .12 %;S期的细胞数由对照组的 0 .0 0 %增加到 6 .6 6 %,处于DNA合成期的细胞数明显增加。结论 :同型半胱氨酸能促进大鼠动脉平滑肌细胞的增殖 ,呈明显的剂量依赖关系。推测 HCY可能通过促进平滑肌细胞的增殖导致动脉粥样硬化的发生。Objective:To observe the effect of Homocysteine(HCY) on the proliferation of the arterial smooth muscle cells(ASMC)of rats.Methods:Through the technique of cell culture in vitro,and by the methods of cell counting,MTT colorimetric analysis ,cell cycle determination with a stream cell instrument,to observe the effect of Homocysteine(HCY)on the proliferation of the arterial smooth muscle cells(ASMC)of rats.Results:HCY could facilitate the proliferation of the ASMCs.After two days of agent acting,the results from cell counting and MTT colorimetric analysis indicated that,compared with those of the control group,the cell number of the HCY group increased by 46.48% and the OD value by the MTT colorimetric analysis increased by 45.69%(P<0.01);As the results from cell cycle determination indicated,by the action of HCY,the cell number in the G 0-G 1 period decreased from 87.22% of the control group to 76.65%,that was,by 12.12%;the cell number in the S period increased from 0.00% of the control group to 6.66%,showing that the cell number in the synthesis period of DNA increased remarkably.Conclusion:HCY can facilitate the proliferation of the ASMCs of rats,and it presents dependence on its dosage remarkably.It should also be indicated that HCY may lead to the occurrence of AS by facilitating the proliferation of the vascular smooth muscle cells.
