速效救心丸对动脉粥样硬化大鼠血脂、主动脉形态及主动脉壁ABCA1表达的影响

摘　　要：[目的]观察速效救心丸对动脉粥样硬化大鼠血脂、透射电镜下主动脉病理形态学及主动脉壁ABCA1的影响,探讨速效救心丸抗动脉粥样硬化(AS)的机制。[方法]采用高脂饮食加一次性大剂量钙负荷方法建立大鼠AS模型。选取60只雄性SD大鼠,随机分为正常组、模型组、速效救心丸低、中、高剂量组及西药对照组,每组各10只。用药组灌服相应剂量的药物,正常组及模型组灌服同等体积的生理盐水,每日1次,连续12周。12周后采血测定血脂,观察主动脉病理形态学改变并测定主动脉壁三磷酸腺苷结合盒转运子A1(ABCA1)的表达。[结果]模型组胆固醇、甘油三酯、低密度脂蛋白和高密度脂蛋白均显著升高(P<0.05),与模型组相比,速效救心丸中、高剂量能降低胆固醇、甘油三酯和低密度脂蛋白,升高高密度脂蛋白水平(P<0.05),并且主动脉病理改变较模型组明显减轻。速效救心丸低、中、高剂量组ABCA1表达均有所上升,与模型组相比,低、中剂量差异没有统计学意义(P>0.05),而高剂量组差异有统计学意义(P<0.05)。[结论]速效救心丸能够明显改善AS大鼠血脂谱,减轻主动脉粥样硬化病理改变。其抗AS、调节脂代谢紊乱的机制部分可能是通过上调ABCA1表达而实现的。[Objective] To observe the effects of Suxiao Jiuxin pills on the blood lipid and the pathologic change in rat of atherosclerosis.[Methods] Experimental atherosclerosis in rats was produced by feeding with atherogenic-diet and vitamin D3 administration. The60 male mature Sprague-Dawlay rats were divided into 6 groups randomly and equally： normal group; AS model group; SXL group; SXM group; SXH group and Astorvastatin group. SX or Astorvastatin was given by gavage from the beginning of the first week to the end of the experiment. At the same time, normal saline was given to the rats of normal group and model group with the same volume and the same way. After 12 weeks, the contents of total cholesterol（TC）, triglyceride（TG）, low density lipoprotein cholesterol（LDL-C） and high density lipoprotein cholesterol（HDL-C） in serum of rats were determined. The pathologic change of artery was observed. [Results] In the model group, TC、TG、LDL and HDL were increased significantly compared with the normal group（P〈0.05）. The levels of TC, TG and LDL were declined and the level of HDL was increased evidently in the SXM, SXH groups（P〈0.05）. Compared with the model group, the pathological changes of aortic ultra structure alleviated obviously in the SXM, SXH and ATO groups. Compared with the model group, the expres-sion of ABCA1 of aortic was not influenced in the SXL, SXM and ATO group（P〉0.05）, while, it was increased in the SXH group（P〈0.05）.[Conclusion] Suxiao Jiuxin pills has obvious effect on decreasing the serum levels of TC, TG, LDL, and increasing the serum of HDL in AS rats. Pathological changes of aorta shows Suxiao Jiuxin pills can ameliorate the atherosclerotic changes. Suxiao Jiuxin pills can in-crease the expression of ABCA1 of aortic.
