摘 要:目的探讨乙肝病毒表面抗原(HBsAg)对Fas介导的人肝癌细胞株细胞毒性及凋亡的影响。方法构建pcDNA3.1-HBsAg重组载体并筛选表达HBsAg的肝癌细胞株,通过CCK-8实验检测HBsAg对Fas激动型抗体(antiFas CH11)介导的HepG2、Huh7、Hep3B细胞毒性的影响;通过TUNEL和AnnexinⅤ实验检测HBsAg对CH11介导的HepG2、Huh7、Hep3B细胞凋亡的影响;自噬抑制剂3-MA预处理后,观察HBsAg对CH11介导的HepG2细胞凋亡的影响。结果 HBsAg以表达量依赖的方式促进CH11介导的HepG2细胞毒性(P<0.05);HBsAg促进CH11介导的HepG2细胞凋亡可以被Fas封闭性抗体(anti-Fas ZB4)完全阻断(P<0.05);过表达mFas受体后HBsAg可促进CH11介导的Huh7和Hep3B细胞毒性及凋亡(P<0.05);CH11介导的HBsAg促凋亡效应可被3-MA抑制(P<0.05)。结论 HBsAg促进Fas介导的肝细胞凋亡依赖于mFas受体,可能与HBsAg诱导自噬机制有关。Objective To discuss the effect of HBsAg on Fas-mediated cytotoxicity and apoptosis of hepatoma cells.Methods pcDNA3.1-HBsAg recombinant vector was constructed and hepatoma cells expressing HBsAg were screened.The effect of HBsAg on Fas-mediated cytotoxicity of HepG2,Huh7 and Hep3 Bcells was tested by CCK-8 assay.The effect of HBsAg on Fas-mediated apoptosis of hepatoma cells was tested by TUNEL and AnnexinⅤ assay.After pretreatment by autophagy inhibitor 3-MA,effect of HBsAg on CH11-mediated HepG2 apoptosis was detected.Results HBsAg promotes CH11-mediated cytotoxicity against HepG2 cells in a dose-dependent manner(P0.05).CH11-mediated apoptosis of HepG2 cells promoted by HBsAg can be completely blocked by Fas blocking antibody ZB4(P0.05).HBsAg promoted CH11-mediated cytotoxicity and apoptosis of Huh7 and Hep3 Bcells after overexpression of mFas receptor(P0.05).CH11-mediated proapoptotic effect by HBsAg was significantly inhibited by 3-MA(P0.05).Conclusion Regulation of Fas-mediated apoptosis of hepatoma cells by HBsAg depends on mFas receptor,which may be related to HBsAg induced autophagy.
