摘 要:目的:观察电针结合天麻素对阿尔茨海默病(Alzheimer disease,AD)模型大鼠海马CA1区乙酰胆碱转移酶(ChAT)和乙酰胆碱酯酶(AChE)表达的影响。方法:SD大鼠随机分为正常对照组、生理盐水组、模型组、电针组、天麻素组、针药结合组,每组10只。腹腔注射D-半乳糖并联合双侧海马注射Aβ1-40制备AD模型大鼠。电针组大鼠电针"百会"、"大椎"、"足三里"穴位,天麻素组大鼠腹腔注射天麻素,针药结合组大鼠腹腔注射天麻素结合电针"百会"、"大椎"、"足三里"穴位,免疫组织化学染色法检测AD大鼠海马CA1区ChAT、AChE的表达。结果:与正常对照组或生理盐水组比较,模型组大鼠海马CA1区ChAT的表达降低,AChE的表达增高(P<0.05);与模型组相比,电针组和天麻素组大鼠海马CA1区ChAT的表达增高,AChE的表达降低(P<0.05);与电针组和天麻素组比较,针药结合组大鼠海马CA1区ChAT的表达上升,AChE的表达降低(P<0.05)。结论:电针结合天麻素可能通过上调大鼠海马CA1区ChAT的表达和抑制AChE的表达而发挥抗AD的作用。Objective: To observe the effects of electro-acupuncture therapy in combination with gastrodin on the expression of acetylcholinesterase( AChE) and acetylcholine transferase( ChAT) in the hippocampal CA1 region of rat models of Alzheimer disease( AD). Methods: SD rats were randomly divided into groups of normal control,normal saline treatment,models,simple electro-acupuncture therapy,simple gastrodin treatment and combined electro-acupuncture with gastrodin( n = 10 for each group). Rat models of AD were prepared by intra-peritoneal injection of D-galactose combined with injection of Aβ1-40 at bilateral hippocampi. Rats in electro-acupuncture group underwent acupuncture at the points of"Baihui","Dazhui"and"Zusanli". Animals in the gastrodin group were given intraperitoneal gastrodin,and those in combined electro-acupuncture with gastrodin group were stimulated at"Baihui","Dazhui"and"Zusanli". Immunohistochemical staining was used to detect the expression of AChE and ChAT in hippocampal CA1 region of AD rats. Results: ChAT expression was significantly down-regulated,and AChE expression was markedly up-regulated in the hippocampal CA1 region of model rats compared to animals in the normal control and saline treatment groups( P〈 0. 05). Significantly increased ChAT,yet decreased AChE expression in the hippocampal CA1 region were seen in rats treated with either simple electro-acupuncture or gastrodin as well as combined electro-acupunctur with gastrodin as compared with the model group or rats treated with simple electro-acupuncture or gastrodin( all P〈 0. 05). Conclusion: Electro-acupuncture in combination with gastrodin may produce effects on AD through up-regulating ChAT expression,yet inhibiting the expression of AChE in hippocampal CA1 region in AD rat models.
