摘  要:目的:建立一种简单、快速、有效的方法测定黄芪中总黄酮含量。方法:在NaNO_3-Al(NO_3)_3-NaOH比色法辅助下,用紫外分光光度法测定黄芪中总黄酮的含量。结果:显色法最佳条件为5%NaNO_3-8%Al(NO_3)_3-5%NaOH,最大吸收波长为512nm,吸光度A与芦丁在0.015~0.6mg/mL范围内呈良好的线性关系(R^2=0.9995),线性方程为A=0.0741c+0.0005,总黄酮的回收率为97.6%,相对偏差(RSD)为2.47%。结论:该法简单准确,易操作,回收率高,可作为黄芪总黄酮含量测定的一种方法。Objective:To establish a simple,rapid and effective method for determination of total flavonoids in Astragalus membranaceus.Methods:The content of total flavonoids in Astragalus membranaceus was determined by UV spectrophotometry with the aid of Na NO3-Al(NO3)3-Na OH colorimetric method.Results:The optimal conditions for chromogenic assay were 5%Na NO3-8%Al(NO3)3-5%Na OH,the maximum absorption wavelength was 512 nm,and the absorbance A was in a good linear relationship with rutin in the range of 0.015-0.6 mg/m L(R^2).=0.9995),the linear equation is A=0.0741c+0.0005,the recovery of total flavonoids was 97.6%,and the relative deviation(RSD)was 2.47%.Conclusion:This method is simple and accurate,easy to operate,and has a high recovery rate.It can be used as a method for the determination of total flavonoids in Scutellaria baicalensis.
