摘  要:目的:利用星点设计-响应曲面法对金钗石斛多糖提取工艺进行优化研究。方法:在单因素试验的基础上,自变量为金钗石斛的提取温度、提取时间、料液比,因变量为多糖得率,通过对自变量各水平的多元化线性回归及二项式拟合,采用星点设计-响应面法选取金钗石斛多糖提取最佳工艺,并进行预测分析。结果:金钗石斛多糖最优提取工艺条件为:提取温度为65℃,料液比为1∶45,提取时间为72 min,提取次数为2次。提取率预测值为7.49%,实际平均提取率为7.57%,相对误差为0.8%。结论:星点设计-响应曲面法优化金钗石斛多糖提取工艺简便合理,稳定性高,精确度高,可预测性较优。Objective: To optimize polysaccharide extraction from Dendrobium nobile by central composite design - response surface methodology. Mothods: On the basis of single factor experiment, the extraction time,extraction temperature, and material - liquid ratio of Dendrobium nobile were taken as independent variables,polysaccharides was taken as the dependent variables, central composite design - response surface methodologywas used to select the optimum extraction process of polysaccharide from Dendrobium nobile through multiplelinear regression and binomial fitting each level of independent variables. Results: The optimum extraction conditions were as follows: the extraction temperature was 65℃, the ratio of material to liquid was 1∶ 45, thee xtraction time was 72 min, with twice extraction. The prediction value of the extraction rate was 7. 49%, andthe actual average extraction rate was 7. 57% with relative error of 0. 8%. Conclusion: The central composite design - response surface methodology is simple and reasonable with high stability and high accuracy to optimize polysaccharide extraction from Dendrobium nobile, and the predictability is better.
