补阳还五汤对舒张性心衰大鼠心肌钙转运蛋白SERCA2a及受磷蛋白的影响

摘　　要：目的运用补阳还五汤对舒张性心衰(diastolic heart failure,DHF)模型大鼠进行干预,观察补阳还五汤对DHF大鼠左心室舒张功能的影响,检测钙转运蛋白肌浆网上的心肌钙转运蛋白(sarcoplasmic reticulum Ca^(2+)-ATPase2a,SERCA2a)及受磷蛋白(phospholamban,PLB)的表达量,初步探寻补阳还五汤治疗DHF的药物干预靶点。方法选用60只雄性SD大鼠,随机分为假手术组15只和模型组45只,利用腹主动脉缩窄法建立舒张性心衰大鼠模型,将造模成功的大鼠随机分为模型组、中药组、西药组,每组11只。其中中药组、西药组灌胃给予相应药物,假手术组、模型组给予等量的去离子水,各组均每日灌胃1次。药物干预8周后,观察药物对超声舒张功能相关指标的影响,运用蛋白免疫印迹法(Western blot)技术检测SERCA2a和PLB的蛋白表达量。结果与模型组相比,中药组大鼠一般状况好转,超声心动图检测二尖瓣环舒张早期运动速度/二尖瓣环舒张晚期运动速度(e’/a’)显著提高(P<0.01)。模型组大鼠心肌中SERCA2a与PLB的蛋白表达量与假手术组相比均有下降(P<0.05),而中药组大鼠心肌中SERCA2a与PLB的蛋白表达量较模型组则升高(P<0.05)。各项结果中药组与西药组差异均无统计学差异(P>0.05)。结论补阳还五汤可改善DHF大鼠左室舒张功能,在一定程度上延缓病情发展,且其的潜在作用靶点可能是心肌钙转运蛋白SERCA2a与PLB。Objective By using Buyang Huanwu decoction to intervene in diastolic heart failure（ DHF） model rats,to observe the effects of Buyang Huanwu decoction on left ventricular diastolic function in DHF rats,to detect the expression of sarcoplasmic reticulum Ca-（2＋）-ATPase2 a（ SERCA2 a） and phospholaban（ PLB）,and to initially explore the drug intervention target of Buyang Huanwu decoction in the treatment of DHF. Methods Sixty male SD rats were randomly divided into sham operation group and model group,DHF model was established by abdominal aortic coarctation,and then the DHF rats were randomly divided into model group,Chinese medicine group and western medicine group,with 11 rats in each group. The Chinese medicine group and the western medicine group were given the corresponding drugs,and the sham operation group and the model group were given the same amount of deionized water. Each group was given intragastric once daily. Drug intervention last for 8 weeks,and then the effect of drug in-tervention on the indexes of ultrasound diastolic function was observed. Western blot was used to detect the protein expression of SERCA2 a and phospholaban PLB on sarcoplasmic reticulum. Results Compared with the model control,the rats in the Buyang Huanwu decoction group improved the general condition.Echocardiographic examination showed that mitral annulus early diastolic velocity/mitral annulus diastolic late motor velocity（ e＇/a＇） was significantly improved（ P〈0. 01）. The protein expression of SERCA2 a and PLB in the myocardium of the model group was decreased compared with the sham group（ P〈0. 05）.The protein expression of SERCA2 a and PLB in the myocardium of the Chinese medicine group was higher than that of the model group（ P〈0. 05）. There was no significant difference between the Chinese medicine group and western medicine group（ P〉0. 05）.
