摘  要:目的探讨逍遥散防治雷公藤致急性肝损伤的可能作用机制。方法 30只SD大鼠随机分为正常组、模型组、逍遥散组,每组10只。逍遥散组大鼠造模前4天灌胃逍遥散6.75 g/(kg·d),正常组和模型组给予等体积蒸馏水,每天1次。第5天模型组和逍遥散组大鼠灌胃雷公藤水煎剂3.75 g/(kg·d)连续4天,建立大鼠急性肝损伤模型。比较各组大鼠肝脏指数及血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)水平,检测肝脏组织B细胞淋巴瘤/白血病-2(Bcl-2)和Bcl-2相关X蛋白(Bax)蛋白表达及天冬氨酸特异性半胱氨酸蛋白酶3(Caspase-3)活性。结果与正常组比较,模型组大鼠的肝脏指数和血清ALT、AST水平明显升高,肝脏组织Bcl-2蛋白表达及Bcl-2/Bax值明显下降,Bax蛋白表达和Caspase-3活性明显升高(P<0.05或P<0.01);与模型组比较,逍遥散组大鼠的肝脏指数和血清ALT、AST水平明显降低,肝脏组织Bcl-2蛋白表达及Bcl-2/Bax值明显升高,Bax蛋白表达和Caspase-3活性明显下降(P<0.05或P<0.01)。结论逍遥散对雷公藤致大鼠急性肝损伤的防控作用可能与其上调肝脏组织Bcl-2蛋白水平和下调Bax蛋白水平、Caspase-3活性,从而改善肝细胞凋亡有关。Objective To investigate the possible mechanism of Xiaoyao Powder( 逍遥散) in preventing and treating acute liver injury induced by Tripterygium Wilfordii( 雷公藤). Methods A total of 30 SD rats were randomly divided into normal group,model group and Xiaoyao Powder group,with 10 rats in each group. In the Xiaoyao Powder group,the rats were given intragastric administration of 6. 75 g/( kg·d) of Xiaoyao Powder for 4 days before modeling,and the normal group and the model group were given an equal volume of distilled water once a day. Rat model of acute liver injury was established in the model group of Day 5 and Xiaoyao Powder group by intragastric administration of Tripterygium wilfordii decoction 3. 75 g/( kg·d) for 4 consecutive days. Liver index and serum alanine aminotransferase( ALT) and aspartate aminotransferase( AST) levels were compared between the groups,and the expression of B cell lymphoma/leukemia-2( Bcl-2) and Bcl-2 related X protein( Bax) protein in the liver tissue and cysteine-containing aspartate-specific proteases 3( Caspase-3) activity were detected. Results Compared with the normal group,the liver index and serum ALT and AST levels in the model group were significantly increased. The expression of Bcl-2 protein and Bcl-2/Bax in liver tissue decreased significantly,and Bax protein expression and Caspase-3 activity increased significantly( P〈0. 05 or P〈0. 01); compared with the model group,liver index and serum ALT and AST levels were significantly decreased in Xiaoyao Powder group,and Bcl-2 protein expression and Bcl-2/Bax values ?? in liver tissue were significantly increased. Bax protein expression and Caspase-3 activity were significantly decreased( P〈0. 05 or P〈0. 01). Conclusion The prevention and control effect of Xiaoyao Powder on acute liver injury induced by Tripterygium wilfordii may be related to the up-regulation of Bcl-2 protein level in liver tissue,down-regulation of Bax protein level and Caspase-3 activity,and thus
