小型猪腹腔镜肝缺血再灌注合并肝部分切除对肝细胞凋亡及相关基因影响的研究

摘　　要：为探究小型猪腹腔镜肝脏缺血再灌注合并肝切除损伤对肝细胞凋亡及凋亡相关基因的影响,将12头巴马小型猪随机分为假手术组和模型组,假手术组仅建立气腹60 min,模型组右半肝缺血60 min后进行左半肝切除。应用TUNEL染色法检测肝细胞凋亡,qRT-PCR法检测凋亡相关基因Bax和Bcl-2 m RNA表达的变化,Caspase-3、Caspase-9活性检测试剂盒检测肝组织中Caspase-3和Caspase-9酶活性。结果显示,模型组Caspase-3和Caspase-9活性、Bax m RNA及细胞凋亡数量于术后即刻、1 d、3 d较假手术组显著升高;模型组Bcl-2 m RNA于术后即刻、1 d、3 d较假手术组显著降低。结果表明,腹腔镜肝脏缺血再灌注合并肝部分切除损伤可导致肝细胞凋亡,其机制可能与上调Bax m RNA和下调Bcl-2 m RNA的表达、Caspase-3和Caspase-9活性增强有关。To investigate the effect of laparoscopic ischemia reperfusion combined with hepatectomy on liver apoptosis and expression of apoptosis genes in miniature pigs,12 Bama miniature pigs were randomly divided into sham operation group and model group. In the sham group,pigs were suffered pneumoperitoneum for 60 min. Pigs in model group were subjected to left hepatectomy after right hepatic ischemia for 60 min. The methods of qRT-PCR,enzyme activity detection kit and TUNEL staining were used to detect the m RNA expression of Bax and Bcl-2,the activities of Caspase-9 and Caspase-3,and cells apoptosis in liver tissues.The result showed that the Caspase-3,Caspase-9,Bax m RNA and cells apoptosis in the model group were significantly increased,while Bcl-2 m RNA were significantly decreased compared with sham group on 0 d,1 d,and 3 d after surgery. These data indicated that laparoscopic ischemia reperfusion combined with hepatectomy induced hepatocyte apoptosis,and its mechanism might be related to the enhancement of Caspase-3 and Caspase-9 activity,up-regulation of Bax m RNA and down-regulation of Bcl-2 m RNA expression in liver tissues.
