牛源A型多杀性巴氏杆菌的生物学特性鉴定与致病基因检测

摘　　要：为研究在田间分离的1株牛源多杀性巴氏杆菌的生物学特性及致病特征,对该菌用平板划线法进行纯化;并利用文献中所提及的该菌分型引物及常见毒力基因引物对其血清型及毒力基因进行PCR检测;利用该菌进行动物致病力回归试验,以石蜡切片的形式观察病理变化;并对其具有较强致病性的原因进行分析。结果显示,该菌株为A型多杀性巴氏杆菌;该菌的半数致死量为25.85 CFU/m L,为强毒株;耐药性无明显变化;23种常见毒力基因中检测到19种。动物回归试验观察结果显示,小鼠出现呼吸急促,精神沉郁,畏寒,眼内出现分泌物等临床症状;病变器官组织学观察结果显示,其主要病变为肺泡充血、炎性渗出,脾淋巴细胞减少及巨噬细胞增多,肝细胞及肾小管上皮细胞广泛性坏死等病变。结果表明,该菌具有较强的毒力,病症久治不愈的原因与毒力因子的分布有关。To study the biological characteristics and pathogenic characteristics of 1 bovine Pasteurella multocida strain which isolated from field cases,the strain was purified by flat line method;the primers of the serotype genes and the common virulence genes of P.multocida which mentioned in the references were used to detect the serotype and the virulence genes by PCR;animal pathogenicity regression experiment was carried out,and the pathological changes were observed in detail in paraffin section form;and the reasons for its high pathogenicity were analyzed.Through detection,it has been confirmed that the pathogenic bacteria isolated from cattle was P.multocida serotype A strain;the LD50 of this strain was 25.85 CFU/m L,which was recognized as highly pathogenic strain;drug sensitivity had no notable change;19 of the 23-common virulence genes were detected.The animal regression experiment showed that mice had shortness of breath,depression,chills,eye secretions and other clinical symptoms.The lesion tissues showed the main pathological changes were alveolar congestion,inflammatoryexudation,spleen lymphopenia and macrophages increase,liver cells and renal tubular epithelial cells with extensive necrosis and other diseases.The results showed that the strong virulence and chronic disease caused by this strain was related to the distribution of its virulence factors.
