2014-2015年度淮安市甲型H3N2流感病毒的流行及血凝素基因特征分析

摘　　要：目的了解淮安地区甲型H3N2流感病毒的流行特征和基因型别。方法收集2014年4月-2015年3月间2家哨点医院采集的流感样病例咽拭子,利用实时荧光RT-PCR方法进行流感病毒的快速检测,对流感病毒核酸阳性的样本进行流感病毒HA基因扩增、测序。利用Lasergene软件进行序列比对及核苷酸同源性分析,并分析血凝素主要的抗原位点,利用Mega6.0软件构建系统进化树,对该时间段内的H3N2病毒流行情况进行统计分析。结果 H3N2亚型流感病毒淮安株的核苷酸同源性为98.9%~99.9%,所收集的淮安株均为H3N2亚型的3C.3a系,在进化关系上与广西株A/Guangxi-Longan/1100/2015和淮安株A/Jiangsu-Qingpu/1660/2015关系较近,H3N2流行主要发生在秋冬季节。结论与疫苗株相比,淮安地区H3N2病毒发生变异,提示疫苗株免疫效果下降。Objective To study the epidemiological characteristic and genotype of influenza A（H3 N2） in Huai ＇an. Methods Influenza virus were collected in two sentinel hospitals from April 2014 to March 2015. Real-time fluorescence was used to detect influenza virus. The HA gene was amplified and sequenced. Sequence alignment and nucleotide homology analysis were analyzed by Lasergene software, and the main antigenic sites of hemagglutinin were analyzed. The phylogenetic tree was constructed by Mega6.0 software. The epidemiological characteristics of H3 N2 virus were analyzed statistically. Results The nucleotide homology of Huai＇an strains was 98.9%-99.9%. The Huai＇an strains were all 3 C.3 a group. Huai＇an strains are closer to strain A/Guangxi-Longan/1100/2015 and Huai＇an strain A/Jiangsu-Qingpu/1660/2015 in evolutional relation.H3 N2 epidemic mainly occurred in the autumn and winter. Conclusion Compared with the vaccine strain, H3 N2 virus in Huai＇an area mutated, suggesting that the immune effect of vaccine strain decreased.
