摘 要:目的观察脾气虚模型大鼠心肌组织中葡萄糖转运体4型(GLUT4)的表达水平及心脏功能的变化。方法通过心脏超声测定脾气虚模型大鼠及四君子丸治疗组心脏功能改变情况,并结合定量PCR和免疫印迹等分子生物学技术检测脾气虚模型组及四君子丸治疗组大鼠心肌组织中GLUT4 mRNA及其蛋白的表达水平。结果脾气虚模型组大鼠心脏功能明显下降,心肌组织中GLUT4 mRNA及其蛋白的表达水平降低,而四君子丸治疗组大鼠心脏功能明显恢复,心肌组织中GLUT4 mRNA及其蛋白的表达水平明显提高,差异均有统计学意义(P<0.05)。结论脾气虚可下调心肌GLUT4 mRNA及其蛋白的表达水平,从而影响葡萄糖的转运,导致大鼠心脏能量代谢障碍及心脏功能异常。对脾气虚证有治疗作用的四君子丸可以恢复心脏功能,提高心肌组织中GLUT4 mRNA及其蛋白的表达水平。Objective To observe the expression level of glucose transporter type 4(GLUT4)and cardiac function in myocardial tissue of rats with spleen-qi deficiency. Methods The changes of cardiac function was measured by echocardiography in spleen-qi deficiency model group and Sijunzi pill treatment group,the expression level of GLUT4 and GLUT4 m RNA inmyocardial tissue of rats were detected by quantitative PCR and western blotting in spleen-qi deficiency model group and Sijunzipill treatment group. Results In spleen-qi deficiency model group,the cardiac function of rats were decreased significantly,the expression level of GLUT4 and GLUT4 m RNA in myocardial tissue were decreased,but in Sijunzi pill treatment group,thecardiac function of rats were obviously recovered,the expression level of GLUT4 and GLUT4 m RNA in myocardial tissue weresignificantly increased,the differences were all statistically significant(P 0.05). Conclusion Spleen-qi deficiency can de-crease the expression level of GLUT4 and GLUT4 m RNA,and affect glucose transportation,lead to cardiac energy metabolismdisorder and cardiac function abnormal.However,Sijunzi pill can recover cardiac function and increase the expression level ofGLUT4 and GLUT4 m RNA in myocardial tissue.
