骨髓间充质干细胞对Ⅱ型胶原诱导性关节炎大鼠的治疗作用

摘　　要：目的探讨骨髓间充质干细胞对Ⅱ型胶原诱导性关节炎(CIA)大鼠的治疗作用。方法将造模成功的大鼠分为模型组、干细胞组、泼尼松组,每组10只,正常组10只。观察大鼠的关节肿胀程度,进行大鼠关节炎指数评分,检测大鼠白细胞介素17(IL-17)、IL-6水平。结果与正常组相比,模型组大鼠关节肿胀程度加重,关节炎指数评分增加,IL-17、IL-6水平升高,差异均有统计学意义(P<0.05);与模型组相比,干细胞组、泼尼松组大鼠关节肿胀程度减轻,关节炎指数评分降低,IL-17、IL-6水平降低,差异均有统计学意义(P<0.05);与干细胞组相比,泼尼松组大鼠关节肿胀程度减轻明显,关节炎指数评分和IL-17、IL-6水平降低显著,差异均有统计学意义(P<0.05)。结论骨髓间充质干细胞可以减轻CIA大鼠关节肿胀程度,降低关节炎指数评分,降低IL-17、IL-6水平,对CIA大鼠具有一定的治疗作用。Objective To study the therapeutic effect of bone marrow mesenchymal stem cells（MSCs）in CIA rats.Methods The 30 SD rats that made the model successfully were divided into model group,stem cell group and prednisone group with 10 rats in each group,in addition,normal group with 10 rats.Then the degree of joints swelling,arthritis index andthe levels of IL-17 and IL-6 in 4 groups were measured,respectively. Results In the model group,the degree of joints swellingwere more serious,the arthritis index and the levels of IL-17 and IL-6 were increased,when compared with the normal group,the differences were statistically significant（P0.05）;however,compared with the model group,the degree of joints swelling,the arthritis index and the levels of IL-17 and IL-6 in the stem cell group and prednisone group were reduced,the differenceswere statistically significant（P0.05）;Compared with the stem cell group,the degree of joints swelling,the arthritis index andthe levels of IL-17 and IL-6 in the prednisone group were significantly decreased,the differences were statistically significant（P0.05）. Conclusion MSCs have a potential therapeutic effect in CIA rats through reducing the degree of joint swelling,decreasing the arthritis index and the levels of IL-17 and IL-6.
