摘  要:目的研究木犀草素对顺铂诱导肝癌HepG2细胞凋亡的增敏作用及机制。方法将HepG2细胞分为空白组、对照组和低、中、高质量浓度实验组。对照组HepG2细胞给予3 mg·L^(-1)的顺铂处理,低、中、高质量浓度实验组HepG2细胞分别以2.5,10,20μg·mL^(-1)木犀草素+3 mg·L^(-1)的顺铂处理,空白组HepG2细胞则以等量生理盐水处理。以四甲基偶氮唑盐(MTT)法检测HepG2细胞增殖抑制情况;以流式细胞术检测HepG2细胞凋亡情况;以蛋白免疫印迹法(WB)检测各组HepG2细胞磷酸化C-jun氨基末端激酶(p-JNK)蛋白表达情况。结果干预24,48,72 h后,实验组HepG2细胞增殖抑制率均高于对照组,且随木犀草素质量浓度增大及作用时间延长呈逐渐升高趋势(P<0.05)。对照组和低、中、高质量浓度实验组HepG2细胞细胞凋亡率分别为(5.23±0.26)%,(11.43±0.67)%,(16.84±1.47)%,(22.63±2.15)%,均显著高于空白组的(2.84±0.31)%,实验组细胞凋亡率高于对照组(P<0.05);随木犀草素质量浓度升高,实验组HepG2细胞凋亡率逐渐升高,细胞增殖指数逐渐下降(P<0.05)。空白组、对照组和低、中、高质量浓度实验组p-JNK蛋白相对表达量为3.68±0.22,0.35±0.03,0.82±0.03,1.23±0.15,2.01±0.19,对照组和低、中、高质量浓度实验组p-JNK蛋白表达量较空白组降低,且实验组均高于对照组(P<0.05)。结论木犀草素通过上调HepG2细胞中p-JNK蛋白表达增加顺铂诱导肝癌HepG2细胞凋亡的敏感性。Objective To investigate apoptosis sensitization and mechanism of luteolin on liver cancer HepG2 cells induced by cisplatin.Methods HepG2 cells were divided into blank group,control group and experimental-L/M/H groups. Cells in control group were treated with 3 mg· L^-1 cisplatin,cells in experimental-L/M/H group were treated with 3 mg · L^-1 cisplatin and 2. 5,10,20 μg · mL^-1 luteolin,blank group was treated with equivalent normal saline. The HepG2 cells proliferation inhibition were detected by Methylthiazolyldiphenyl-tetrazolium( MTT) method,the HepG2 cells apoptosis were detected by flow cytometry,phosphorylated C-jun N terminal kinase( p-JNK) protein expression of HepG-2 cells in each group were detected by Western blot( WB). Results After 24,48 and 72 h intervention,the HepG2 cells proliferation inhibition in experimental group were all higher than those incontrol group and gradually increased with the increase of luteolin concentration and the prolongation of action time( P〈0. 05). The HepG2 cells apoptotic rates of control group and experimental-L/M/H groups were( 5. 23 ± 0. 26)%,( 11. 43 ± 0. 67) %,( 16. 84 ± 1. 47) %,( 22. 63 ± 2. 15) %,higher than( 2. 84 ± 0. 31) % in blank group,the apoptosis rates in experimental group were higher than that in control group( P〈0. 05). With the concentration of luteolin increasing,the HepG2 cells apoptotic rate of experimental group was gradually increased and the cell proliferation index decreased gradually( P〈0. 05). The relative expression levels of p-JNK protein in HepG2 cells in blank group,control group and experimental-L/M/H groups were 3. 68 ± 0. 22,0. 35 ± 0. 03,0. 82 ± 0. 03,1. 23 ± 0. 15,2. 01 ± 0. 19. The p-JNK protein expression in control group and experimental-L/M/H groups were lower than that in blank group and experimental group were higher than control group( P〈0. 05). Conclusion Luteolin could increase cisplatin-induced apoptosis sensitization of HepG2 cells by up-regulating p-JN
