来曲唑对子宫内膜异位症大鼠模型的影响研究

摘　　要：目的研究来曲唑对子宫内膜异位症大鼠模型的保护作用及对异位内膜细胞凋亡相关基因表达的影响。方法将32只建模成功的子宫内膜异位症雌性SD大鼠随机分为模型组及实验组,各16只。实验组灌胃0.25 mg·kg^(-1)来曲唑,模型组则灌胃等量生理盐水,每天1次,2组均连续干预3周。比较2组大鼠病灶体积及病理学形态变化;以末端转移酶介导的缺口末端标记法(TUNEL)检测2组异位内膜细胞凋亡情况;以反转录聚合酶链式反应(RT-PCR)检测2组异位内膜组织中相关基因表达情况。结果模型组及实验组大鼠治疗后病灶体积分别为(158.39±11.23),(75.14±10.65)mm^3,与干预前相比差异有统计学意义(P<0.01)。实验组异位组织细胞凋亡率为(5.64±1.12)%,显著高于模型组的(2.06±1.01)%(P<0.01)。模型组细胞色素芳香化酶P450(P450arom)、环氧合酶-2(COX-2)、B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白基因(Bax)mRNA相对表达量分别为0.43±0.02,0.49±0.04,0.47±0.03,0.48±0.02;实验组分别为0.21±0.03,0.35±0.02,0.33±0.05,0.61±0.03,差异均有统计学意义(均P<0.01)。结论来曲唑对子宫内膜异位症大鼠具有显著的保护作用,其作用机制可能与促进异位组织细胞凋亡,下调P450 arom、COX-2及抗凋亡基因Bcl-2表达相关。Objective To explore the protective effect of letrozole on the endometriosis rats and analyze the influence on the expression of apoptosis-related genes of ectopic tissue cells. Methods Thirty-two cases successful modeled endometriosis female SD rats were divided randomly into model group and test group,16 rats in each group. Test group was gavaged 0. 25 mg·kg^-1 letrozole,and model group was gavaged with equivalent normal saline,1 time a day. Two groups were intervened for 3 weeks. The lesion volume and pathology morphology changes of two groups were compared; the apoptosis of two groups were detected by terminal dexynucleotidyl transferase（ Td T）-mediated d UTP nick end labeling（ TUNEL）; the expression of apoptosis-related genes of ectopic tissue cells in 2 groups were detected by reverse transcription-polymerase chain reaction（ RT-PCR）. Results The lesion volume of model group and test group after intervened were（ 158. 39 ± 11. 23）,（ 75. 14 ± 10. 65） mm^3,had significant difference with those before intervented（ P〈0. 01）. The apoptosis rate of ectopic tissue cells in test group and model group were（ 5. 64 ± 1. 12） %,（ 2. 06 ± 1. 01） %,with significant difference（ P〈0. 01）. The relative expression of cytochromeP450 aromatase（ P450 arom）,cyclooxygenase-2（ COX-2）,B cell lymphoma/lewkmia-2（ Bcl-2）,Bcl-2 associated X protein（ Bax） mRNA in model group were 0. 43 ± 0. 02,0. 49 ± 0. 04,0. 47 ± 0. 03,0. 48 ± 0. 02; had significant difference with those in test group,which were 0. 21 ± 0. 03,0. 35 ± 0. 02,0. 33 ± 0. 05,0. 61 ± 0. 03（ P〈0. 01）. Conclusion Letrozole has remarkable protective effect on the endometriosis rats,the action mechanism maybe related to promote the apoptosis of ectopic tissue cells,down-regulation of the expression of P450 arom,COX-2 and antiapoptotic gene Bcl-2.
