未控制哮喘与ADRB2基因位点多态性关系的分析

摘　　要：目的探讨β2肾上腺素能受体基因(β2-adrenergicreceptor,ADRB2)rs1042718、rs2895795及rs12654778位点基因多态性与未控制哮喘的相关性。方法选取2015年12月—2016年11月我院确诊为支气管哮规范化治疗半年后未控制者30例为病例组,部分控制者30例为阴性对照组,同期我院健康体检者30例为正常对照组,采用Sequenom Mass AR R AY?SN P检测法和基质辅助激光解吸附电离飞行时间质谱分析质谱技术对患者血液中上述三个位点进行多态性检测。结果 rs12654778位点存在GG/GA/AA三种基因型,其基因型的分布频率在三组无统计学意义(GG/GA/AA),且其基因型及等位基因的分布频率在三组间比较差异无统计学意义(P=0.554/0.297/0.819);rs2895795位点TT/AT/AA三种基因型,在三组间比较差异无统计学意义(P=0.105/0.104/0.730);rs1042718位点基因型三组间比较差异无统计学意义(P=0.141/0.051/0.846)。结论 ADRB2基因多态性不是未控制哮喘发病的遗传决定因素,这可能与种族差异、单倍型有关。Objective To investigate the correlation between gene polymorphisms of β 2-adrenergicreceptor（ADRB2） gene including rs12654718、rs2895795 and rs12654778 and refractory asthma. Method From Dec 2015 to Dec 2016, patients being diagnosed as bronchial asthma and being treated with β 2 adrenergic beta-agonists therapy in our hospital for half a year,were divided into two groups. 30 cases was into group without being cured, and 30 cases was into partly cured group. There were other 30 cases of healthy people being divided into control group. All subjects were genotyped by Sequenom MassARRAY？SNP assay and matrix-assisted laser desorption/ionization time of flight（MALDI-TOF） method. Result The rs12654718 Gene loci had three genotypes：GG/GA/AA. The distribution of the genotype frequency in the three groups had no statistical significance,The distribution of genotype and allele frequency among three groups was no statistically significant difference（P =0.554/0.297/0.819）..The rs2895795 Gene loci had three genotypes：TT/AT/AA. And there was no statistically significant difference between three groups（P =0.105/0.104/0.730）.There was no statistically significant difference about the rs1042718 genotype between three groups（P =0.105/0.104/0.730）.Conclusion ADRB2 genetic polymorphism was not associated with uncontrolled asthma.This may be related to racial differences and, haplotype of gene.
