摘 要:目的观察银翘柴桂Ⅱ号方对流感病毒FM1感染小鼠的死亡保护作用,探讨其对TLR7/My D88/NF-κB信号通路的影响。方法制备流感病毒性肺炎小鼠模型,观察银翘柴桂Ⅱ号方对模型小鼠的死亡保护作用;ELISA法测定小鼠血清TNF-α、IL-1β、IL-6水平;荧光定量PCR和WB法检测小鼠肺组织TLR7、My D88、NF-κB m RNA和蛋白水平的表达量。结果银翘柴桂Ⅱ号方低、中、高剂量组均能提高模型小鼠的死亡保护率,降低模型小鼠血清中TNF-α、IL-1β、IL-6水平,下调模型小鼠肺组织中TLR7、My D88、NF-κB m RNA和蛋白水平的表达(P<0.05,P<0.01)。结论银翘柴桂Ⅱ号方可以提高流感病毒性肺炎小鼠的死亡保护率,表现出抗流感病毒作用,该作用通过抑制由病毒激活的TLR7/My D88/NF-κB信号通路实现。Objective To observe the protective effect of Yinqiao Chai Gui prescription Ⅱ on death of influenza virus FM1 infected mice,and to investigate its effect on TLR7/My D88/NF-κB signaling pathway. Methods Mice were infected with FM1 virus to prepare pneumonia models caused by influenza virus,and the death protective effect of Yinqiao Chai Gui prescription Ⅱ was observed. The serum levels of TNF-α,IL-1β,IL-6 in infected mice were measured by ELISA. Real-time RT-PCR and Western Blot assay were used to detect the expression levels of TLR7,My D88,NF-κB m RNA and protein in lung tissue. Results Yinqiao Chai Gui prescription Ⅱ low,medium and high dose groups significantly increased the death protection rate,reduced the levels of serum TNF-α,IL-1β,IL-6 in FM1 infected mice. The m RNA and protein expression levels of TLR7, My D88, NF-κB in lung tissue of FM1 infected mice were significantly decreased by treatment of Yinqiao Chai Gui prescription Ⅱ low,medium and high doses. Conclusion Yinqiao Chai Gui prescription Ⅱ improved the death protection rate in virus infected mice;the underlying mechanism of antiviral effect might be associated with TLR7/My D88/NF-κB signaling pathway.
