摘 要:目的:观察自拟"益气复元汤"对溃疡性结肠炎大鼠Th1/Th2细胞平衡的影响。方法:采用2,4,6-三硝基苯磺酸(2,4,6-TNBS)/乙醇法复制UC大鼠模型,给予柳氮磺吡啶溶液及11.92、17.16、23.84g/kg剂量益气复元汤水煎液干预14天,观察各组大鼠一般情况、记录疾病活动指数(DAI),进行大鼠结肠黏膜损伤指数(CMDI)及结肠组织学损伤评分(HI);检测外周血淋巴细胞亚群及细胞因子TNF-α、IL-4和INF-γ的含量。结果:与正常组比较,模型组DAI、CMDI、HI和INF-γ、TNF-α含量以及CD4+、CD4+/CD8+淋巴细胞百分比显著升高,IL-4含量和CD8+、CD4+CD25+/CD4+淋巴细胞百分比显著降低;柳氮磺吡啶及益气复元汤各剂量组,可显著降低模型大鼠DAI、CMDI、HI和INF-γ、TNF-α、CD4+的含量以及CD4+/CD8+淋巴细胞百分比,增加IL-4、CD8+的表达以及CD4+CD25+/CD4+淋巴细胞百分比。结论:益气复元汤可抑制TNBS/乙醇诱导的溃疡性结肠炎大鼠模型的炎症,可能通过减少血清INF-γ表达、上调IL-4表达,从而调节Th1/Th2细胞平衡,调节肠道免疫,抑制肠道炎症,促进溃疡愈合。Objective: To investigate the effect of the herb compound Yiqifuyuan decoction on Th1/Th2 polarization in the rat model of ulcerative colitis. Methods: Ulcerative colitis in the rat was induced by 2,4,6-trinitrobenzenesulfonicacid(TNBS). All rats were randomly divided into six groups: the control group,the model group,sulfasalazine oral treatment group,and high,moderate,and low doses of Yiqifuyuan decoction groups(10 rats in each group). Disease activity index(DAI),Colon mucosal damage index(CMDI) and histological injury(HI) were evaluated after 14 days,and contents of TNF-α,INF-γand IL-4 in serum were detected. Results: Compared with the model group,DAI,CMDI,HI,and contents of TNF-α and INF-γ in decoction groups and sulfasalazine oral treatment group were effectively reduced(P〈0. 05). The serum IL-4 content in the model group was significantly lower(P〈0. 05) than that in the control group,while those in decoction groups were significantly improved(P〈0. 05). Conclusion: The research suggests that Yiqifuyuan decoction is effective and has significant effects on Pro-inflammatory cytokine TNF-α and INF-γ,and inflammatory cytokine suppression IL-4 in rats with ulcerative colitis induced by TNBS by regulating the Th1/Th2 type cells balance.
