摘 要:目的探究诸葛菜种子的化学成分。方法采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、ODS柱色谱、重结晶等方法进行分离纯化,利用化合物的理化性质及NMR等波谱学技术,结合ESI-MS等方法对分离所得化合物进行结构鉴定。采用MTT法测定分离得到的化合物体外抗肿瘤活性。结果从诸葛菜种子的70%乙醇提取物正丁醇萃取部位分离得到12个化合物,分别鉴定为5,7,3′,5′-四羟基二氢黄酮(1),染料木素(2),金合欢素-7-O-β-D-葡萄糖苷(3),落叶松树脂醇(4),落叶松树脂醇-9-O-β-D-吡喃型葡萄糖苷(5),异落叶松树脂醇-9-O-β-D-吡喃型葡萄糖苷(6),薯蓣皂苷(7),纤细薯蓣皂苷(8),胡萝卜苷(9),β-谷甾醇-3-O-β-D-吡喃葡萄糖苷-20-O-棕榈酸酯(10),长寿花葡萄糖苷(11),hypargyreusolide B(12)。结论除化合物1和9外,其余化合物均为首次从诸葛菜植物中分离得到。化合物4、6、7和8对人肝癌细胞株(Hep G2)和人非小细胞肺癌细胞株(A549)具有显著的细胞毒性,IC_(50)值为0.88~20.33μmol/L。Objective To study the chemical constituents of the seeds of Orychophragmus violaceus. Methods The compounds were isolated by silica gel,Sephadex LH-20,ODS column chromatography and recrystallization. Their structures were identified based on the chemical evidence and spectroscopic analysis,combined with the ESI-MS method. Compounds obtained were assayed for their cytotoxicities using the MTT assay in vitro. Results Twelve compounds were isolated from the n-Bu OH part of 70%ethanol extract of the seeds of O. violaceus. These compounds were identified as 5,7,3′,5′-tetrahydroxyflavanone(1),geristein(2),acactin-7-O-β-D-glucoside(3),lariciresinol(4),lariciresinol-9-O-β-D-glucopyranoside(5),isolariciresinol-9-O-β-D-glucopyranoside(6),dioscin(7),gracillin(8),daucosterol(9),β-sitosteryl-3-O-β-D-glucopyranoside-20-O-palmitate(10),roseoside(11),and hypargyreusolide B(12),respectively. All the compounds were assayed for their cytotoxicities in vitro by MTT. Conclusion These compounds,except 1 and 9,are isolated from the plant for the first time. Compounds 4,6,7 and 8 showed significant cytotoxicities on human cancer A549 and Hep G2 cells,with IC50 values ranging from 0.88 to 20.33 μmol/L.
