摘  要:目的:观察芪乌调脂颗粒对动脉粥样硬化大鼠炎症介质、基质金属蛋白酶、血管内皮生长因子(VEGF)的影响,探索芪乌调脂颗粒稳定动脉粥样硬化斑块的作用机制。方法:将Wistar大鼠80只随机分为空白组、模型组、芪乌调脂颗粒组、阿托伐他汀组、芪乌调脂颗粒+阿托伐他汀组,除空白组外均给予高脂饮食喂饲4周,病理提示造模成功后按组别分别给予芪乌调脂颗粒、阿托伐他汀钙,连续给药8周,测定超敏C反应蛋白(hs-CRP)水平,光镜下观察主动脉组织病理形态学改变,并利用免疫组织化学法观察VEGF及基质金属蛋白酶9(MMP-9)在主动脉的表达情况。结果:1)与空白组比较,模型组大鼠血清hs-CRP水平明显上升,主动脉组织中MMP-9、VEGF阳性表达升高,差异有统计学意义(P<0.01);2)与模型组相比,阿托伐他汀组、芪乌调脂颗粒+阿托伐他汀组hs-CRP、MMP-9阳性反应降低,芪乌调脂颗粒+阿托伐他汀组VEGF阳性表达降低,差异均有统计学意义(P<0.05);3)与阿托伐他汀组相比,芪乌调脂颗粒+阿托伐他汀组hs-CRP、MMP-9、VEGF降低差异有统计学意义(P<0.05)。结论:芪乌调脂颗粒协同阿托伐他汀钙可通过降低炎症介质、基质金属蛋白酶、血管内皮生长因子起到稳定动脉粥样斑块的作用。Abstract Objective: To observe the effects of QiWu TiaoZhi granules on inflammatory mediators, matrix metalloproteinase (MMP) and vascular endothelial growth factor (VEGF) of atherosclerotic rats, and explore its mechanism of QiWu TiaoZhi granules stabilizing atheroslcerotic plaque. Methods: Eighty rats were randomized into the blank group, the model group, QiWu TiaoZhi granules group, atorvastatin group and QiWu TiaoZhi granules+atorvastatin group, except the blank group, the other rats were fed with high fat food for four weeks, after pathological results showed that the models were successfully prepared, they were given QiWu TiaoZhi granules and atorvastatin according to the grouping respectively, they were administered for eight weeks consecutively, the expressions of MMP-9 and VEGF in the aorta were observed by using immunohistochemistry, hs-CRP was detected, and histopathological changes of the aorta was observed under light microscope. Results: 1) compared with the blank group, the levels of hs-CRP increased obviously in the model group, positive expressions of MMP-9 and VEGF in the aorta raised, and the difference had statistical meaning (P〈0.01); 2) compared with the model group, positive expressions of hs-CRP and MMP-9 decreased in atorvastatin group, QiWu TiaoZhi granules+atorvastatin group, positive expressions of VEGF lowered in QiWu TiaoZhi granules+atorvastatin group, and all the difference showed statistical meaning (P〈0.05); 3) compared with atorvastatin group, the levels of hs-CRP, MMP-9 and VEGF in QiWu TiaoZhi granules+atorvastatin group decreased, and the difference showed statistical meaning (P〈0.05). Conclusion: QiWu TiaoZhi granules and atorvastatin could stabilize atheroslcerotic plaque through decreasing MMP and VEGF
