摘  要:[目的]探讨野玫瑰根水提取物(ARR)对大鼠离体血管功能的影响及其机制.[方法]采用大鼠离体血管功能实验装置,分别记录血管张力变化.观察累加质量浓度ARR(0.1~100.0mg/L)对去氧肾上腺素(PE,1.0μmol/L)预收缩有内皮和去内皮血管环的作用;采用不同阻断剂预处理后观察ARR对血管环张力改变的影响.[结果]ARR剂量依赖性地舒张PE预收缩的内皮完整的大鼠胸主动脉环.与内皮完整血管组比较,内皮去除、左旋硝基精氨酸甲酯及ODQ预处理可显著抑制ARR对血管的舒张效应(P<0.05).维拉帕米及地尔硫卓预处理可部分抑制ARR对内皮完整血管的舒张效应(P<0.05),而吲哚美辛、格列本脲及TEA预处理未见明显抑制ARR对内皮完整血管的舒张作用(P>0.05).[结论]ARR具有质量浓度依赖性的血管舒张作用,其机制认为可能与内皮依赖性NO-cGMP途径有关系.OBJECTIVE To explore the effects of aqueous extract of Rosa davurica Pall.radix(ARR)on the function of rat isolated vessels and its mechanism.METHODS The experimental apparatus of rat isolated vascular function was used to record the change of vascular tension.The effects of ARR,cumulative mass concentration 0.1-100.0 mg/L,on vascular ring with or without intact endothelium which preshrinked by phenylephrine(PE,1.0μmol/L)were observed.The effects of ARR on the change of vascular ring tension were observed after pretreatment with different blockers.RESULTS ARR dose-dependently relaxed phenylephrine preshrinked rat aortic ring with intact endothelium.As compared with the blood vessel group with intact endothelium,endothelial removal,NG-nitro-L-arginine methyl ester and ODQ pretreatment could significantly inhibit the diastolic effects of ARR on blood vessels(P〈0.05).The pretreatment of verapamil and diltiazem could partially inhibit the diastolic effects of ARR on endothelial intact vessels(P〈0.05),while indomethacin,glibenclamide and TEA pretreatment did not significantly inhibit the diastolic function of ARR in endothelial intact vessels(P〉0.05).CONCLUSION ARR had concentration-dependent vasodilation effect,its mechanism might be related to the endothelial dependent NO-cGMP pathway.
