UPLC-MS法同时测定古汉养生精口服液中5种化学成分

摘　　要：目的建立超高效液相色谱-串联质谱法同时测定古汉养生精口服液中淫羊藿苷、黄芪甲苷、人参皂苷Rb1、Re和Rg1的含量。方法采用Waters Acquity UPLC C18色谱柱(2.1mm×50mm,1.7μm);以乙腈-0.1%甲酸水溶液(15∶85)为流动相进行洗脱,流速0.2mL·min^(-1),以电喷雾离子源多反应监测(MRM)方式负离子模式检测离子对m/z 721.3→513.40(淫羊藿苷),正离子模式分别检测离子对m/z 807.3→627.30(黄芪甲苷)、m/z 1131.4→365.46(人参皂苷Rb1)、m/z 969.5→789.38(人参皂苷Re)、m/z 823.5→643.54(人参皂苷Rg1)。结果淫羊藿苷、黄芪甲苷、人参皂苷Rb1、Re和Rg1分别在1860.0~37200.0ng·mL^(-1)、172.6~3451.8ng·mL^(-1)、95.8~1915.2ng·mL^(-1)、89.4~1787.0ng·mL^(-1)、98.4~1968.0ng·mL^(-1)浓度范围内线性关系良好,回收率均在99.31%~100.75%之间,RSD均小于1.55%(n=6)结论此法简单、快速、准确可靠,可用于古汉养生精口服液的质量控制。OBJECTIVE To develop an UPLC-MS method for the simultaneous determination of Icariin, Astra- galoside IV, Ginsenosides Rb1, Re, Rgl in GuHanYangShengJing Liquid. METHODS The chromatographic separa- tion was carried out on a Waters Acquity UPLC C18 column（2. 1 mmx 50mm, 1.7 μm） with the mobile phase consis- ting of acetonitrile-0. 1% formic acid with isometric elution at a flow rate of 0. 2mL · min-1. Icariin（ m/z 721.3→513.40 ）, AstragalosideIV （ m/z 807.3→627.30）, Ginsenosides Rb1 （ m/z 1131.4→365.46 ）, Ginsenosides Re （ m/z 969.5→789.38） and Ginsenosides Rg1 （m/z 823.5→643.54 ） were detected by tandem mass spectrometry under the positive and negative ions mode with the electrospray ionization source and in the multiple reaction monitoring （MRM） mode. RESULTS Excellent chromatographic separation was achieved and the range for linear correlation of Icariin,Astragaloside IV,Ginsenosides Rb1, Ginsenosides Re and Ginsenosides Rgl were 1860. 0- 37200. 0ng · mL-1,172. 6 - 3451.8ng ·mL-1 , 95. 8 - 1915.2ng · mL-1, 89. 4 - 1787.0ng · mL-I and 98. 4 - 1968.0ng · mL-1,respectively. The average recoveries of the four components were between 99.31% and 100. 75 （ RSD ≤ 1.55% ）. CONCLUSION The established method is simple,rapid,sensitive and accurate,which can be applied to quality control of GuHanYangShengJing Liquid.
