双波长HPLC法测定痢泻灵片中穿心莲内酯和脱水穿心莲内酯的含量

摘　　要：目的建立用双波长HPLC法测定痢泻灵片中穿心莲内酯和脱水穿心莲内酯含量的方法。方法采用双波长高效液相色谱法,色谱柱:C18反相色谱柱,流动相:甲醇-水(55∶45),检测波长:穿心莲内酯225nm,脱水穿心莲内酯254nm。结果穿心莲内酯在0.04~0.81μg范围内,其峰面积与浓度呈良好的线形关系Y=2.2302×105X-10446(相关系数r=0.9999),平均回收率为100.0%,RSD为1.4%(n=6)。脱水穿心莲内酯在0.04~0.80μg范围内,其峰面积与浓度呈良好的线形关系Y=1.7716×105X-7643(其中相关系数r=0.9999),平均回收率为100.7%,RSD为1.5%(n=6)结论本方法快速、准确、重复性好,可用于测定痢泻灵片中穿心莲内酯和脱水穿心莲内酯的含量。OBJECTIVE To establish a HPLC with dual-wavelength method for determination of andrographol- ide and dehyandrographolide in LiXieLing tablets. METHODS By high performance liquid chromatography with dual-wavelength ,chromatography column was C18 ,mobile phase was methanol-water （55：45）,detection wavelength was 225nm of andrographolide and 254nm of dehyandrographolide. RESULTS The andrographolide in the range of 0. 1 - 1.6ug, the peak area and concentration had good linear relationship with Y = 5. 0958 - 10^5X - 4347 （ r = 0. 9999） ,the average recovery was 100. 0%, RSD was 1.4% （n = 6）. The dehyandrographolide in the range of 0. 2 2.41xg,the peak area and concentration had good linear relationship with Y =5. 3681 x 10^5X ＋24918（r =0. 9999） ,the average recovery was 100. 7% ,RSD was 1.5% （n =6） CONCLUSION The method is rapid,accurate and reproduci- ble. It can be used for the determination of andrographolide and dehyandrographolide in LiXieLing tablets.
