摘 要:本文首先合成了含还原敏感双硫键的N,N'-双(丙烯酰)胱胺(CBA),并以此为交联剂、以α-甲基丙烯酸为主单体、通过改变交联剂及单体的投料组成制备了三组还原敏感型纳米凝胶。该纳米凝胶具备较高的药物包载能力;包封率与载药量均随CBA含量的增加而增大。体外药物控释实验证实纳米凝胶具备较好的还原敏感性,可在癌细胞内的还原环境下迅速释放模型抗癌药物阿霉素;且释药速度随CBA含量的增加而明显加快。该纳米药物载体系统具备良好且可控的药物控释能力,具备一定的研究价值。In this paper, we synthesized redox-sensitive cystamine-N,N'-bisacrylamide(CBA) with disulfide bond, which was then used as the crosslinking agent to fabricate poly(α-methylacrylic acid)(PMAA) nanogels. By adjusting the amount of CBA and PMAA, three kinds of nanogels were obtained. The in vitro controlled drug release experiments demonstrated that the nanogles had excellent redox-sensitivity and could release model anti-cancer drug doxorubicin hydrochloride inside the cancer cells due to the reducing environment intracellular. And the rate of drug releasing also increased with the increasing of CBA content in the nanogles. With the outstanding controlled drug release behavior, the nanogel based drug delivery systems have great potential for anti-cancer researches.
