摘　　要：目的进行小鼠急性毒性和大鼠长期毒性实验,了解其毒性情况,为临床实验提供依据。方法急性毒性:小鼠40只常规饲养,实验前禁食12 h,按体质量随机分为雌、雄对照及雌、雄给药组4组。各给药组以最大可灌饲浓度(0.5 g/ml),按0.3 ml/10g体质量灌胃给药,2次/d,对照组给予等体积的蒸馏水,连续12 d。观察动物的不良反应和死亡情况,记录体质量增长情况,计算最大给药量。长期毒性:大鼠160只,按体质量随机分为正常对照组和给药大(15 g/kg/d)、中(7.5 g/kg/d)、小(3.75 g/kg/d)剂量组,雌雄各半,连续6个月给药,2次/d。对照组给予蒸馏水。给药后3、6个月及停药后4周解剖动物。结果最大给药剂量(30g/kg/d)下未发现小鼠有明显的毒副反应,总用药量相当于人临床用药量的100倍;连续6个月给药期间各剂量组小鼠一般情况、进食量、外周血象、生化指标、心电、脏器指数无明显影响;主要脏器无明显病理性改变。结论太空养心丸未出现明显的毒副反应,提示该药临床长期用药是安全的。Objective To investigate the toxicity and provide evidence for clinical experiment,we conduct the acute toxicity test of mice and the chronic toxicity test of rats. Methods The acute toxicity test of mice： 40 mice were randomly divided into two groups,control group and administration group,with 20 mice in each group and half male and half female. The mice in the administration group were fed by Taikong Yangxin pills at the maxim dose of 30 g/kg/d. The mice in the control group were fed with equal volumes of distilled water. We observed the adverse reaction,the death status,and the growth of weight of the mice. The chronic toxicity test of rats： 160 rats were randomly divided into four groups fed with distilled water,and herbal（ 15 g/kg/d,7. 5 g/kg/d,3. 75 g/kg/d respectively） for six months. The rats were dissected after 3 and 6 months of administrating and drug withdrawal for 4 weeks. Results There were not obvious adverse reactions on mice. There were not obvious changes on the rats in the general conditions,food intake,peripheral hemogram,biochemical indexes,viscera index and cardiac electrical activity.There were not significant pathological changes of the main viscera. Conclusion There were not obvious toxicity or adverse reactions in the acute toxicity test of mice or the chronic toxicity test of rats,indicating it is safe to take Taikong Yangxin pills clinically for long duration.
