摘 要:目的:比较棕色脂肪干细胞(brown adipose stem cells,BADSCs)和白色脂肪干细胞(white adipose stem cells,WADSCs)的体外成脂能力。方法:分离SD大鼠脂肪组织,获得BADSCs和WADSCs。显微镜观察细胞形态、流式鉴定细胞表面抗原标记物,并成脂诱导两种细胞。诱导后油红O染色显示形成的脂滴。RT-PCR检测成脂相关基因的表达。结果:BADSCs和WADSCs都具有成脂分化能力;与BADSCs相比,WADSCs脂滴形成相对较快且数量较多,RT-PCR结果显示PPAR、C/EBPβ成脂标志基因表达也相对较高。结论:BADSCs和WADSCs都具有成脂分化能力。相比较而言,WADSCs有较强的成脂分化能力。Objective: To compare the adipogenic capacity of brown adipose derived stem cells( BADSCs) and white adipose stem cells( WADSCs). Methods: SD rat adipose tissue was isolated and BADSCs and WADSCs were obtained. Microscopic observation of cell morphology,flow cytometric identification of cell surface antigen markers,and adipogenic induction of both cells. Oil Red-O staining after induction showed the formation of lipid droplets. RT-PCR detected the expression of adipogenic related genes. Results: Both BADSCs and WADSCs had adipogenic differentiation ability. Compared with BADSCs,the formation of droplets in WADSCs was relatively rapid and the number was large. RT-PCR results showed that the PPAR and C/EBPβ adipogenic marker genes were also relatively high. Conclusion: Both BADSCs and WADSCs have adipogenic differentiation ability. In comparison,WADSCs have strong adipogenic differentiation.
