摘 要:目的研究黄芩素对人结肠癌细胞生长及肿瘤抑制基因PTEN(phosphatase and tensin homologue,PTEN)、Akt及p-Akt表达的影响。方法以人结肠癌SW620细胞为研究对象,通过Ed U和MTT实验方法检测0、25、50、75μmol/L浓度的黄芩素对肿瘤细胞生长及侵袭力的影响;通过Western blot检测黄芩素对肿瘤细胞PTEN、Akt及p-Akt蛋白的表达。结果 Ed U实验结果显示,浓度为50、75μmol/L的黄芩素能够显著抑制SW620细胞的DN A复制活性(P<0.05);MTT实验表明黄芩素能够抑制SW620的细胞增生,呈剂量、时间依赖性;Transwell小室迁移实验显示50、75μmol/L浓度的黄芩素能抑制SW620细胞的侵袭迁移能力(P<0.05);WB实验结果显示,50、75μmol/L浓度的黄芩素能够上调PTEN的表达,下调p-Akt的表达(P<0.05)。结论黄芩素能够经PTEN/Akt途径抑制结肠癌肿瘤细胞的生长和侵袭力。Objective To study the influence of scutellaria baicalensis georgi on cell growth of colon cancer,phosphatase and tensin homologue(PTEN),Akt and p-Akt expression.Methods SW620 cell of colon cancer was selected as the research subject.The influence of scutellaria baicalensis georgi with the concentrations of 0,25,50 and 75 μmol/L on the growth and invasive power of the cancer cell were tested with the experimental methods of EdU and MTT.Western blot was used to test the influence of scutellaria baicalensis georgi on PTEN of the cancer cell,Akt and p-Akt protein expression.Results The experimental result of EdU showed scutellaria baicalensis georgi with the concentrations of 50 and 75 μmol/L could significantly inhibit the DNA replication activity of SW620 cell( P〈0.05).The MTT experiment showed scutellaria baicalensis georgi could inhibit the proliferation of SW620 cell,showing the dosage and time dependence.Transwell cell migration experiment showed scutellaria baicalensis georgi with the concentrations of 50 and 75 μmol/L could inhibit the invasive ability of SW620 cell to migrate( P〈0.05).The result of WB experiment showed scutellaria baicalensis georgi with the concentrations of 50 and 75 μmol/L could increase the expression of PTEN and decrease p-Akt expression( P〈0.05).Conclusion Scutellaria baicalensis georgi could inhibit the growth and invasive power of cell of colon cancer through PTEN/Akt.
