摘  要:目的:探讨斑螯素对卵巢癌细胞中Smad3、转化生长因子-β1(transforming growth factor-β,TGF-β1)、氨基端激酶(cJun N-terminal kinase,c-JUN)表达的影响,探究其抗肿瘤的作用机制。方法:将卵巢癌细胞进行常规复苏培养,选取对数生长期细胞分为两组,观察组加入斑螯素培养,对照组加入体积分数0.1%的DMSO培养。于培养12 h、24 h两个时间点进行检测。采用实时荧光定量PCR法检测(real-time PCR,RT-PCR)卵巢癌细胞中Smad3、TGF-β1、c-JUN的mRNA水平。采用酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测卵巢癌细胞中Smad3、TGF-β1、c-JUN的蛋白水平。分析Smad3、TGF-β1、c-JUN的mRNA与其蛋白水平的相关性,以及斑螯素对Smad3、TGF-β1、c-JUN表达的影响。结果:观察组24 h时,卵巢癌细胞中Smad3、TGF-β1、c-JUN的mRNA及蛋白水平均显著低于12 h(P<0.05),其细胞增殖抑制率显著高于12 h(P<0.01)。对照组24 h时,卵巢癌细胞中Smad3、TGF-β1、c-JUN的mRNA及蛋白水平均显著高于12 h(P<0.05),其细胞增殖抑制率与12 h比较,差异无统计学意义(P>0.05);在培养12 h、24 h时,观察组卵巢癌细胞中Smad3、TGF-β1、c-JUN的mRNA及蛋白水平分别低于对照组12 h、24 h的水平(P<0.05),其细胞增殖抑制率显著高于对照组(P<0.01);各组不同时间点卵巢癌细胞中Smad3、TGF-β1、c-JUN的mRNA分别与其蛋白表达呈正相关(r>0)。结论:斑螯素抗肿瘤、抑制细胞的增殖作用与卵巢癌细胞Smad3、TGF-β1、c-JUN的表达有密切关系。Objective:To study the effect of canechelin on the expression of Smad3,transforming growth factor-β1( TGF-β1) and cJun N-terminal kinase( c-JUN) in ovarian cancer cells and to explore its anti-tumor mechanism of action. Methods:Ovarian cancer cells were subjected to conventional resuscitation culture. The logarithmic phase cells were divided into two groups. The observation group was added with canelirain in culture,and the control group was incubated with DMSO with a volume fraction of 0. 1%. The test was performed at two time points:12 h and 24 h. The mRNA levels of Smad3,TGF-β1,and c-JUN in ovarian cancer cells were determined by real-time fluorescent quantitative PCR. Enzyme-linked immunosorbent assay( ELISA) was used to detect the protein levels of Smad3,TGF-β1 and c-JUN in ovarian cancer cells. The correlations between mRNA and protein levels of Smad3,TGF-β1,c-JUN,and the effect of canelirain on the expression of Smad3,TGF-β1,and c-JUN were analyzed. Results:The mRNA and protein levels of Smad3,TGF-β1 and c-JUN in ovarian cancer cells in 24 h of the observation group were significantly lower than that in 12 h( P〈0. 05) and the inhibitory rate of cell proliferation was significantly higher than that of 12 h( P〈0. 01). The mRNA and protein levels of Smad3,TGF-β1,and c-JUN in ovarian cancer cells were significantly higher than those in control group at 12 h( P〈0. 05),and the cell proliferation inhibition rate was not statistically different from that at 12 h( P〈0. 05). The mRNA and protein levels of Smad3,TGF-β1 and c-JUN in the ovarian cancer cells in the observation group were lower than those in the control group at 12 h and 24 h( P〈0. 05),and the cell proliferation inhibition rate was significantly higher in the observation group at 12 h and 24 h. The mRNA levels of Smad3,TGF-β1,and c-JUN in ovarian cancer cells at each time point were positively correlated with their protein expression( r〉0). Conclusion:The anti-cancer effect of canelirai
