摘  要:目的观察化合物M-286对糖基化终产物(advanced glycosylation end product,AGE)诱导视网膜血管内皮细胞(retinal microvascular endothelial cell,RMEC)凋亡的影响。方法体外培养RMEC,以不同浓度的化合物M-286培养液作用于AGE处理的RMEC,MTT法检测RMEC存活率,流式细胞术检测凋亡细胞比例及细胞内ROS含量。结果不同浓度的化合物M-286可在不同程度上保护AGE处理后的RMEC,与AGE组比较,能够明显提高细胞存活率(P<0.05)。与空白对照组比较,AGE处理24 h后RMEC凋亡比例明显升高;不同浓度的化合物M-286处理后细胞凋亡比例均有所下降,与AGE组比较,差异具有统计学意义(P<0.05);AGE处理24 h后,ROS水平明显升高,化合物M-286可降低AGE处理后细胞内ROS水平,与AGEs组比较,差异具有统计学意义(P<0.05)。结论化合物M-286可通过抑制AGE诱导RMEC中ROS的产生,减少RMEC凋亡。Objective To study the effects of compound M-286 on the apoptosis of retinal microvascular endothelial cells(RMECs) induced by advanced glycation end products(AGEs) in rats. Methods The retinal microvascular endothelial cells were cultured in vitro and treated with AGEs, then were exposed to various concentrations of compound M-286. The viability of RMECs was measured by MTT, and the percentage of apoptotic cells and intracellular ROS were detected by flow cytometry. Results M-286 of different concentrations showed protective effect on RMECs dealt with AGEs, and significantly increased the cell survival rate(P〈 0.05) compared with that of AGE group. In contrast with the control group, the apoptosis rate of the RMECs significantly increased after being dealt with AGEs for 24 h; the percentage of apoptotic cells decreased after being treated with different concentrations of compound M-286. Compared with AGE group, the difference was statistically significant(P〈 0.05); AGEs markedly elevated the level of ROS in the RMECs, and M-286 reduced the production of ROS in the AGEs-induced RMECs compared with that of AGE group, the difference was statistically significant(P 〈0.05). Conclusion Compound M-286 can reduce the RMEC apoptosis by inhibiting AGEs-induced ROS production.
