摘 要:目的研究大鼠合用雷公藤多苷片和缬沙坦后,雷公藤多苷片对缬沙坦药代动力学的影响。方法 16只雄性SD大鼠,随机分为单独给药组和联合给药组两组,其中单独给药组灌胃缬沙坦(20 mg/kg),联合给药组灌胃雷公藤多苷片(10 mg/kg)和缬沙坦(20 mg/kg)。不同时间点取血并采用LC-MS测定缬沙坦的血药浓度,计算药代动力学参数。结果与单独给药组相比,联合给药组缬沙坦的最大血药浓度[(1.57±0.32)vs(1.01±0.28)μg/m L]和血浆曲线下面积[(10.01±0.58)vs(6.82±0.77)mg·h/L]明显增加(P<0.05),说明缬沙坦的代谢受到明显抑制。结论雷公藤多苷片与缬沙坦联用之后,会对缬沙坦的药代动力学产生明显的影响,本研究对缬沙坦和雷公藤多苷片联合用药具有一定的指导作用,可以避免临床药物不良相互作用的发生。Objective To study the effect of tripterysium glycosides tablets on the pharmacokinetics of valsartan in rats combined with tripterysium glycosides tablets and valsartan. Methods 16 male Sprague-Dawley rats were randomly divided into two groups: the single administration group and the combined administration group. In the single administration group, the rats were given intragastric administration of valsartan(20 mg/kg) and the combined administered group was given intragastric administration of tripterysium glycosides tablets(10 mg/kg) and valsartan(20 mg/kg). Blood samples were taken at different time points and plasma concentrations of valsartan were determined by LC-MS. Pharmacokinetic parameters were calculated. Results The maximum plasma concentration of valsartan was [(1.57 ±0.32) vs(1.01±0.28)μg/m L] and the plasma area under the curve was [(10.01 ±0.58) vs(6.82±0.77)mg·h/L], which were significantly increased in the combined administration group combined with the single administration group(P〈0.05), indicating that valsartan metabolism was significantly inhibited. Conclusion Tripterysium glycosides tablets combined with valsartan has a significant effect on the pharmacokinetics of valsartan. This study plays a guiding role in the combined administration of valsartan and tripterygium glycosides tablets, which can avoid the occurrence of clinical adverse drug interactions.
