3T3-E1细胞增殖分化过程中雷洛昔芬对内质网应激的影响

摘　　要：目的探讨选择性雌激素受体调节剂(selective estrogen receptor modulator,SERM)雷洛昔芬对3T3-E1细胞增殖分化的作用及其机制。方法用不同浓度的雷洛昔芬(10-9mol/L、10-8mol/L、10-7mol/L)处理培养3T3-E1细胞,另设空白对照组,24 h后检测各指标。CCK8法检测细胞增殖,使用碱性磷酸酶测定试剂盒测定碱性磷酸酶(alkaline phosphatase,ALP)活力,实时荧光定量聚合酶链式反应(real time quantitative polymerase chain reaction,q RT-PCR)检测细胞内IRE-1、ATF6、e IF2α的表达。结果雷洛昔芬与对照组相比较可促进3T3-E1细胞增殖(P<0.05),随着雷洛昔芬浓度的增高,细胞增殖程度升高(P<0.05),高浓度的雷洛昔芬(10-7mol/L)促增殖能力最强;雷洛昔芬不同浓度处理后各组ALP活力增加(P<0.05),而且高浓度雷洛昔芬(10-7mol/L)升高ALP活力较明显;IRE-1、ATF6及e IF2α的表达均升高(P<0.05)。结论雷洛昔芬可能通过上调IRE-1、ATF6及e IF2α的表达促进3T3-E1细胞的增殖分化。Objective To explore the role and mechanism of selective estrogen receptor modulator-raloxifene on the proliferation and differentiation of the 3T3-E1 cells.Methods 3T3-E1 cells were cultured with different concentrations of raloxifene （10^-9 mol/L, 10 ^-8mol/L and 10 ^-7mol/L）, and a blank control group was also established.Indicators were measured after 24 hours. Cell proliferation was detected with cell counting kit -8 （CCK8）, ALP activity was determined using the ALP kit, and the expression of IRE-1, ATF6 and eIF2αwere measured by the qRT-PCR.Results Compared with the control group, raloxifene can promote the proliferation of 3T3-E1 cells （P〈0.05）.With the increase of raloxifene concentration, the proliferation of the 3T3-E1 cells increased （P〈0.05）, with the highest concentration of raloxifene （10 ^-7mol/L） had the strongest effect.The ALP activity of the 3T3-E1 cells increased after treatment with raloxifene, and the effect was most evident at the highest concentration of raloxifene.The mRNA expression of IRE-1,ATF6 and eIF2αincreased in with raloxifene treatment （P〈0.05）.Conclusion Raloxifene could promote the proliferation and differentiation of 3T3-E1 cells through up-regulating the mRNA expression of IRE-1, ATF6 and eIF2α.
