摘 要:试验以慈竹种子为材料,探讨不同激素浓度配比对愈伤组织诱导和增殖的影响.结果表明:诱导愈伤组织的最佳培养基为MS+3.0 mg/L 2,4-D+0.5 mg/L 6-BA,可诱导出浅绿色,质地松软颗粒状,不规则团状的I型胚性愈伤组织,诱导率达到38.25%.最佳增殖培养基为MS+2.5 mg/L KT+3.0 mg/L 6-BA,增殖率为76.08%,增殖系数为2.10.The seeds of Neosinocalamus affinis used as mainly explant in this experiment,the callus induction and proliferation was studied on the different kind ratio of hormone concentrations.The results showed that the best medium of callus induction was MS + 3.0 mg/L 2,4-D + 0.5 mg/L 6-BA,and obtained Type I callus which color was reseda,had granulariy irregular crumb granulated surface with a soft texture.The Induction rate was 38.25 %.The 2,4-D and 6-BA interaction was played a leading role in the callus induction.The best proliferation medium was MS with KT 2.5 mg/L and 6-BA3.0 mg/L,which the increment rate was 76.08 %.The proliferation coefficient was 2.10.The KT and 6-BA interaction was played a improment role in the proliferation.
