摘 要:目的研究血管紧张素Ⅱ(AngⅡ)诱导的心脏重构的机制和筛选潜在治疗靶点。方法大鼠心脏成纤维细胞(CFSs)加入10nMAngⅡ后培养12h,对照组未加入AngⅡ。从2组样品中提取RNA后,检测细胞基因表达水平。使用R语言中的limma软件包筛选两组样本间的差异表达基因(DEGs),并通过富集分析探讨它们的功能和参与的信号通路。此外,通过蛋白质-蛋白质相互作用(PPI)网络构建揭示它们在蛋白水平的关系。结果共筛选了126个上调基因和140个下调差异表达基因。根据富集分析发现:其中的ACACB,IL1B,IL1A,NOS2和MMP3与免疫反应、血管生成的调控、超氧代谢过程和羧酸结合生物过程高度相关。PPI网络构建显示:ACACB和MPP3是两个主要节点。结论 ACACB,IL1B,IL1A,NOS2和MMP3这五种基因,可能是AngⅡ诱导心脏重构的重要候选标志物。Objective To reveal the mechanism of Angiotensin Ⅱ(Ang Ⅱ)-induced cardiac remodeling and to identify the potential therapeutic targets. Methods The rat cardiac fibroblasts(CFSs) were cultured with 10 n M Ang Ⅱ for 12 h while those used as control were without Ang Ⅱ; after RNA extraction from the 2 kinds of the samples, RNA-sequencing was performed to detect gene expressions; the differentially expressed genes(DEGs) were identified using limma package in R, and their function and pathways were explored through enrichment analysis, and, their potential relationships at protein level were revealed by protein-protein interaction(PPI) network. Results In total, 126 up-regulated and140 down-regulated DEGs were identified;enrichment analysis indicated that ACACB, IL1 B, IL1 A, NOS2 and MMP3 were highly associated with immune response,regulation of angiogenesis, superoxide metabolic process and carboxylic acid binding biological processes. Among them; ACACB and MPP3 were also two predominant nodes in the PPI network. Conclusions The 5 genes: ACACB, IL1 A, NOS2, IL1 B and MMP3, might be used as candidate markers for the prevention of Ang II-induced cardiac remodeling.
