浙江省一规模猪场副猪嗜血杆菌的分离鉴定

摘　　要：2017年12月,浙江省杭州市萧山区某规模猪场保育舍猪群出现高热(41.5℃)、咳嗽、腹式呼吸、关节肿胀伴有跛行和被毛粗乱等临床症状。为确诊疾病,明确病因,剖检2头40日龄病死猪,用接种环刮取肺脏病料接种血平板,再用金黄色葡萄球菌作交叉划线,于37℃5%CO2条件下培养24 h。结果显示:病料培养血平板上生长出大量半透明、不溶血小菌落,菌落直径为0.5~1 mm;靠近金黄色葡萄球菌菌苔的菌落较大,随着与葡萄球菌生长线距离的增加而变小或不生长,呈"卫星现象"。挑取可疑菌落进行纯化培养,将纯培养物制成菌悬液,以DNA纯化试剂盒提取DNA并进行PCR检测。扩增产物电泳后,被检菌株电泳道出现一条1 090 bp条带,表明该菌株为副猪嗜血杆菌。In December 2017,some pigs appeared high fever（41.5℃）,cough,abdominal breathing,swelling of joints,claudication,rough hair and other symptoms in the nursery houses of a large-scale swine farm in Xiaoshan District of Hangzhou City,Zhejiang Province.In order to identify the cause,2 died pigs of 40 days old were dissected.Using the inoculation ring,the samples of lung tissue were inoculated on blood plate,and the staphylococcus aureus was also used to make cross-lines on the plate.At last,the plate was cultured for 24 hours under the condition of 37 ℃ 5% CO2.Results showed that a large number of semi-transparent and non-hemolytic micro colonies grew on the blood plate,the colony diameter was 0.5 to 1 mm.Besides,the colonies near the lawns of staphylococcus aureus were larger,and the size of these colonies reduced with the increasing distance from the colonies to the staphylococcal growth line,showing＂satellite phenomenon＂.Then the suspected colonies were selected and cultured,and the pure cultures were made into bacterial suspensions.Using the DNA purification kit,the DNA of the suspensions were extracted and amplified by PCR,and a electrophoretic band of 1 090 bp was got,which showed that the bacterial strain was Haemofhilus parasuis.
