摘  要:以黄瓜白粉病菌为试验材料,比较分析了改良CTAB法、SDS法和真菌试剂盒法对黄瓜白粉病菌基因组DNA提取的效果。结果表明,CTAB法提取的黄瓜白粉菌基因组DNA在纯度(R=A_(260 nm)/A_(280 nm))和产量上均优于SDS法和真菌试剂盒法,且杂质少。CTAB法提取的黄瓜白粉菌基因组DNA产率为204.3μg/g,而SDS法和真菌试剂盒法分别为147.7、117.7μg/g,且方法产率之间差异极显著。采用CTAB法提取的DNA纯度较高,为1.969 6;SDS法和真菌试剂盒法提取的DND纯度较低,分别为1.832 2和1.507 9。With cucumber powdery mildew bacteria as test material,the effects of modified CTAB method,SDS method and fungal kit method on genomic DNA extraction of cucumber powdery mildew bacteria were compared and analyzed. The results show that the genomic DNA extracted by CTAB method was superior to SDS method and fungus kit method in purity(R=A(260 nm)/A(280 nm))and yield,and had less impurities. The DNA production rate of cucumber powdery mildew extracted by CTAB method was 204.3 μg/g,while SDS method and fungi kit method were 147.7 μg/g and 117.7 μg/g,and variance of the yield from different methods was very significant. The high purity of DNA extracted by CTAB method was 1.969 6,while the low purity of SDS method and fungus kit method were 1.832 2 and 1.507 9,respectively.
