摘 要:采用ISSR分子标记技术,对采自大连地区3个黄精野生种群(共79个个体)和5个多花黄精野生种群(共146个个体)进行了遗传多样性分析和比较.12个ISSR引物在黄精和多花黄精种群分别扩增出137条和161条条带.其中,黄精种群多态位点比率(PPB)、Nei’s指数(h)和Shannon信息指数(I)分别为86.86%、0.279 7和0.423 4,其遗传分化系数(G_(st))和基因流(N_m)分别为0.398 2和0.755 6;多花黄精种群PPB、h、I分别为94.41%、0.3398、0.5059,其G_(st)和N_m分别为0.3084和1.1215.研究结果表明,黄精和多花黄精天然种群均具有较高的遗传多样性,且黄精种群遗传多样性明显低于多花黄精.黄精各种群的遗传多样性偏低,种群间较大的遗传差异导致其物种水平的遗传多样性较高.The genetic diversity of 3 natural populations of Polygonatum sibiricum(79 individuals)and 5 natural populations of Polygonatum cyrtonema Hua(146 individuals),which were collected from Dalian,was analyzed and compared by ISSR molecular markers technology.The number of bands amplified by 12 ISSR primers was 137 and 161,respectively in P.sibiricumand P.cyrtonema Hua populations.In P.sibiricum populations,the percentage of polymorphic bands,Nei's index,Shannon information index was 86.86%,0.2797,0.4234,its genetic differentiation coefficient,gene flow was 0.3982,0.7556,which was 94.41%,0.3398,0.5059 and 0.3084,1.1215 in P.cyrtonema Hua populations.This study suggests that both P.sibiricumand P.cyrtonema Hua natural populations have rich genetic diversity,the former was significantly lower than the latter.The genetic diversity was low in each population of P.sibiricumand the large genetic differences among populations contributed to the rich genetic diversity at species level.
