摘  要:菰属(Zizania Trucz.)为水稻的近缘属,保留了大量驯化作物所丢失的优异性状,是扩大和丰富育种基因库的理想材料。从24对菰SSR分子标记中筛选出7对标记,对来自东北三省的5个居群,共计98个野生菰样本进行了遗传多样性和遗传结构分析。同时,详细阐述了POPGENE、STRUCTURE、ARLEQUIN、MEGA及MVSP等统计分析软件的数据录入格式、软件操作方法,为相关领域的研究者提供完整可操作的参考依据。各软件分析结果表明:东北地区野生菰具有较为丰富的遗传多样性(PPB=71.88%、Ao=1.719、Ae=1.332、He=0.201、I=0.313,Gst=0.543);各野生菰居群间的遗传相似性系数和遗传距离分别介于0.605 5~0.955 6和0.045 4~0.501 6;东北地区野生菰的遗传变异主要存在于居群间;5个野生菰居群的遗传结构可以大致分为3组,5个居群不存在明显的亲缘地理关系。该研究结果不仅有助于加深对我国东北地区野生菰资源遗传多样性和遗传结构的认识,同时也为东北野生菰种质资源保护提供一定理论依据。Zizania Latifolia(Griseb.) Trucz. is an aquatic herbaceous plant of Oryzeae tribe,which is a close genera of rice and an important carrier of genes controlling excellent agronomic traits of significant importance for rice breeding.In this study,seven out of 24 pairs of SSR markers were used for genetic diversity and genetic structure analysis of 5 wild populations from Northeast China. POPGENE,STRUCTURE,ARLEQUIN,MEGA and MVSP,were introduced for its data format and usage methods.This work showed that Z.latifolia resources in Northeast China had a rich genetic diversity. The results of POPGENE analysis showed that there was rich genetic diversity in the wild species(PPB = 71.88 %、Ao = 1.719、Ae = 1.332、He = 0.201、I = 0.313,Gst = 0.543) and the genetic similarity coefficient and genetic distance were between 0.6055 - 0.9556 and 0.0454 - 0.5016,respectively.Based on molecular variation analysis by ARLEQUIN,it was showed that the genetic variation of the 5 wild populations mainly exists among populations while low degree of variation was tested within populations.For the genetic structure analysis,5 populations were divided into three groups.Cluster analysis showed that the 5 wild populations of Z.latifolia could be classified into two groups(one group was DL,and the other was SY,QQHE,JMS,TH). There was no significant association between the phylogenetic relationship and the geographic locations in the Z.latifolia populations.This study deepens our understanding about the genetic evolution of Z.latifolia in Northeast China and at the same time provides certain theoretical basis for protection of this wild rice germplasm resource.
