摘 要:目的考察不同剂量脉复生对血栓闭塞性脉管炎(TAO)模型大鼠血管内皮炎症的影响。方法通过注射月桂酸钠建立TAO模型,实验分为空白组、模型组、脉复生低剂量组(2.5 g/kg)、中剂量组(5 g/kg)、高剂量组(10 g/kg),给药结束对大鼠TAO病变进行分级评分;血液黏度仪测定全血黏度和血浆黏度;Western blot和Realtime PCR法检测血管内皮炎症指标细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)和白介素-6(IL-6)的蛋白表达和转录水平。结果与模型组比较,不同剂量组脉复生均可有效缓解模型大鼠病变(P<0.05),降低全血黏度和血浆黏度(P<0.05,P<0.01),降低并抑制ICAM-1、VCAM-1和IL-6蛋白表达和转录水平(P<0.05,P<0.01)。结论脉复生能减轻TAO大鼠血管内皮炎症反应。Objective To study the effect of Maifusheng on vascular endothelial inflammation in rats with thromboangiitis obliterans( TAO). Methods TAO rat model was established by injection of sodium laurate.Rats were divided into the blank group, the model group, Maifusheng low-dose group( 2. 5 g/kg),Maifusheng medium-dose group( 5 g/kg) and Maifusheng high-dose group( 10 g/kg). TAO pathological changes were assessed by grading score. The whole blood viscosity and plasma viscosity were determined by blood viscosimeter. The expression and transcription level of intercellular adhesion molecule-1( ICAM-1),vascular cell adhesion molecule-1( VCAM-1) and interleukin-6( IL-6) were detected by Western blot and Real-time PCR. Results Compared with the model group, different doses of Maifusheng effectively alleviated the pathological changes in TAO rats( P〈0.05),decreased the whole blood viscosity and plasma viscosity( P〈0.05,P〈0.01),and reduced the expression and transcriptional of ICAM-1,VCAM-1 and IL-6( P〈0.05,P〈0.01). Conclusion Maifusheng could reduce the vascular endothelial inflammation in TAO rats.
