摘  要:【目的】了解唐家河国家级自然保护区中华蜜蜂的遗传多样性,为进一步保护和利用唐家河中华蜜蜂资源提供科学依据。【方法】以基因片段mt DNA t RNAleu^COⅡ的非编码区作为分子标记,对唐家河各个地区共37个中蜂样本进行序列测定及相关分析。【结果】唐家河中蜂的核苷酸多样度(Pi)为0.030 34,单倍型多样性(Hd)为0.839±0.041;共发现14个单倍型,主体单倍型TJH2和TJH4分别占总样本数的29.73%和27.03%。此外本研究首次报道了3个新发现的单倍型。【结论】唐家河中蜂具有丰富的遗传多样性,其单倍型的分布与地理、气候等因素有较大关系。【Objective】The purpose of this study was to understand the genetic diversity and provides the scientific basis for protecting and utilizing the resources of Apis cerana cerana in Tangjiahe National Nature Reserve,Sichuan,China.【Method】The mitochondrial DNA t RNAleu~CO Ⅱ sequences of 37 samples from Tangjiahe National Nature Reserve were determined and analyzed.【Result】It was shown that the nucleotide polymorphism(Pi) was 0.030 34 and the haplotype diversity(Hd) was 0.839±0.041;14 haplotypes were found in 37 honeybee colonies. TJH2(29.73%) and TJH4(27.03%) were the main haplotypes found in the 37 honeybee colonies. Moreover,three new haplotypes were found in Tangjiahe National Nature Reserve. 【Conclusion】Tangjiahe National Nature Reserve has abundant genetic diversity and the haplotype distribution,which was connected with geography,climate and other factors. It provides important theoretical basis for the protection and reasonable using of the genetic resources of Apis cerana cerana in Tangjiahe National Nature Reserve.
