摘  要:目的:采用HPLC技术,建立降糖保健品中违禁添加的格列美脲、格列吡嗪、格列本脲、盐酸吡格列酮、瑞格列奈、格列齐特、格列喹酮、盐酸二甲双胍及盐酸苯乙双胍等9种降糖化学药物的检测方法。方法:采用TIANHE Kromasil C18柱(250 mm×4.6 mm,5μm),甲醇-0.1%磷酸二氢钠溶液(用磷酸调节pH值至3.0±0.05)(70∶30)为流动相,流速为1 ml/min,紫外检测波长为233 nm,检测分析格列类及其他类药物;采用TIANHE Kromasil C18柱(250 mm×4.6 mm,5μm),甲醇-离子对试液(辛烷磺酸钠0.585 6 g,加三乙胺1.4 ml,加水至1 000 ml,用磷酸调节pH值至3.5±0.05)(55∶45)为流动相,流速为1 ml/min,紫外检测波长为233 nm,检测分析双胍类药物。结果:该方法分离度好,线性相关性好(r>0.999),检出限为0.40~0.86μg/ml,定量限为1.2~2.6μg/ml,平均回收率为98.8%~104.3%。结论:该方法快速、准确、灵敏度高,能对降糖类中药及保健品中非法添加化学药品进行检测。Objective: To develop an HPLC method to determine nine chemicals with hypoglycemic activity which were illegally added into a health product. Methods: The separation was performed on TIANHE Kromasil C18 column. The mobile phase was made up of methnol-0. 1% sodium dihydrogen phosphate(V∶ V = 70∶ 30) and methnol-ion pair reagent(V∶ V = 55∶ 45) respectively; detection wavelength was set at 233 nm. The flow rate were 1 ml/min. Results: Nine linear calibration curves were obtained with r 0. 999. The limits of detection(LOD) and quantity(LOQ) were 0. 40 - 0. 86 μg/ml and 1. 2 - 2. 6 μg/ml,respectively. The recoveries covered from 98. 8% till 104. 3%. Conclusion: The method is convenient,rapid and accurate and can be used for detecting illegally added hypoglycemic chemicals in health products.
